兔视网膜中央静脉阻塞模型中小胶质细胞CD40和铁蛋白的表达 被引量:4

摘要 目的分析兔视网膜中央静脉阻塞模型中小胶质细胞CD40和铁蛋白表达的变化,探讨其发生机制。方法将30只有色兔随机分为空白组、模型组,以激光动力学方法造模,行眼底照相和眼底荧光血管造影法检查,分别于1 d、3 d、7 d、14 d和28 d对两组兔视网膜及视神经标本采用免疫组织化学方法检测CD40和铁蛋白表达水平。40倍光镜照相,测定灰度值和积分光密度(IOD),对测得数据进行统计学分析。结果小胶质细胞CD40的表达结果 :3 d时空白组和模型组视网膜标本灰度值分别为193.42±9.72和86.03±5.12,差异有统计学意义(P<0.05);3 d时空白组和模型组视神经标本灰度值分别为196.26±10.78和85.91±5.01,差异有统计学意义(P<0.05);3 d时空白组和模型组视网膜标本IOD分别为105.36±7.58和4479.32±307.22,差异有统计学意义(P<0.05);3 d时空白组和模型组视神经标本IOD分别为54.63±8.79和10 436.42±1306.83,差异有统计学意义(P<0.05)。小胶质细胞铁蛋白的表达结果 :3 d时空白组和模型组视网膜标本的IOD分别为161.96±35.37和1295.59±124.71,差异有统计学意义(P<0.05)。结论视网膜中央静脉阻塞可以引起视网膜及视神经小胶质细胞的活化,免疫组织化学染色显示小胶质细胞CD40和铁蛋白表达均为阳性结果 。 Objective To analyze the change of the microglia cells CD40 and fer- ritin expression in the rabbit model of central retinal vein occlusion, and explore its mechanism. Methods Thirty colored rabbits were divided randomly into blank group and model group. The central retinal vein occlusion models were established with laser dynamics method, and then examined with fundus photography and FFA. The expres- sion of CD40 and ferritin in retina and optic nerve at 1 day,3 days,7 days,14 days and 28 days were observed by immunohlstochemistry. Photography was made with 40 times optical microscope, the gray value and IOD were examined. The data were analyzed by statistical software. Results The results of microglia CD40 expression:At 3 days,the gray values in retinal specimens of blank group and model group were 193.42 ± 9.72 and 86.03 ± 5.12, respectively, the difference was statistically significant (P 〈 0.05 ) ; At 3 days,the gray values in optic nerve specimens of blank group and model group were 196.26 ± 10.78 and 85.91 ± 5.01, respectively, the difference was statistically significant (P 〈 0.05 ) ; At 3 days, IOD in retinal specimens of blank group and model group were 105.36 ± 7.58 and 4479.32 ± 307.22,respectively,the difference was statistically signifi- cant (P 〈 0.05 ) ; At 3 days, IOD in optic nerve specimens of blank group and model group were 54.53 ± 8.79 and 10 436.42 ± 1306.83 ,respectively,the difference was statis- tically significant(P 〈0.05). The results of microglia ferritin expression:At 3 days,IOD in retinal specimens of blank group and model group were 161. 96 ± 35. 37 and 1295.59 ± 124.71, respectively, the difference was statistically significant ( P 〈 0.05 ). Conclusion Central retinal vein occlusion can activate the microglia cells in retina and optic nerve. Microglia cells CD40 and Ferritin expression are positive in immunohis- tochemical study.
