摘要
目的观察沙棘对老年大鼠血浆核磁共振(NMR)谱图的影响,以探讨沙棘干预衰老的机制。方法将SD大鼠分为青年组(4月龄,n=10)、老年对照组(24月龄,n=12)及老年干预组(24月龄,n=11)。收集各组大鼠血浆进行1H-NMR检测,应用Hotelling's T2和趋势过滤方法分析各组血浆中代谢物的变化。结果与青年组比较,老年对照组发现与衰老相关的谱峰有28个,鉴定出16种代谢物,这些代谢物主要涉及到糖代谢、氨基酸代谢和脂类代谢。与老年对照组比较,老年干预组中被沙棘逆转的且已鉴定的代谢物有葡萄糖、乳酸、丙酮酸、甘氨酸、谷氨酰胺、亚油酸、棕榈酸、油酸、丙酮和甘油10种,沙棘能明显地改变衰老进程中的代谢物轨迹。结论大鼠衰老进程中存在糖代谢、氨基酸代谢和脂类代谢紊乱,且在不同程度上均可被沙棘逆转,表明沙棘延缓衰老的机制可能与其逆转代谢紊乱有关。
AIM To observe the effects of Hippophae rhamnoides L on blood plasma nuclear magnetic resonance spectra of aged rats in order to study the role of Hippophae rhamnoides L in anti-aging and mechanisms. METHODS SD rats were indivied into 3 groups: 4-month-old young group (n = 10), 24-month-old elderly group (n = 12) and 24- month elderly plus Hippophae rhamnoides L intervention group (n = 11 ). Rats blood plasma was collected and detected by using ^1H-NMR. The alteration of metabolities was analyzed by the method of Hotelling's T^2 and trend filtering algo- rithm. RESULTS Compared with the 4-month-old young group, aging-related 28 peaks were found and 16 kinds of metabolites were identified in the 24-month-old elderly group, which metabolites were mainly related to glucose metabolism, amino acid metabolism and lipid metabolism. Compared with the 24-month-old elderly group, 10 kinds of metabolites, which were reversed by Hippophae rhamnoides L and identified in the 24-month elderly plus Hippophae rhamnoides L intervention group, were glucose, lactic acid, pyruvic acid, glycine, glntamine acid, linoleic -acid, palmitic acid, oleic acid , acetone and glycerol, which indicated that the aging-related metabolic profiles were signifi- cantly reversed by Hippophae rhamnoides L. CONCLUSION There are disorders of glucose metabolism, amino acid metabolism and lipid metabolism during aging process, which can be reversed by Hippophae rhamnoides L, indicating that the mechanism of anti-aging effect of Hippophae rhamnoides L may be related to its reversal of metabolic disorder.
