顺铂联合HSV-tk自杀基因对肝癌细胞HepG2的生长抑制作用 被引量:1

摘要 目的研究HSV-tk自杀基因联合顺铂对人Hep G2细胞株的杀伤作用。方法使用脂质体携带p AFP-TK-IRES2-EGFP质粒转染Hep G2细胞;Hep G2细胞在转染HSV-TK基因后,继续添加顺铂,用CCK-8试剂检测HSV-tk基因联合顺铂对Hep G2细胞杀伤效果。结果重组质粒p AFP-tk-IRES2-EGFP成功转染Hep G2肝癌细胞;HSV-tk自杀基因联合顺铂对Hep G2细胞的杀伤作用大于单独使用顺铂或HSV-tk自杀基因(P<0.05)。结论实验成功评价了顺铂联合HSV-tk自杀基因对Hep G2肝癌细胞株的杀伤效果,为肝癌的临床治疗方法提供了实验依据。 Objective To investigate the killing effect of HSV-tk suicide gene combined with cisplatin in human Hep G2 cell line. Methods The p AFP-TK-IRES2-EGFP plasmid was carried by liposome to transfect Hep G2 cells. After HSV-tk gene was transfected by Hep G2 cells, cisplatin was added and then CCK-8 reagent was used to determine the killing effects of HSV-tk gene combined with cisplatin in Hep G2 cells. Results Recombinant plasmid p AFP-TK-IRES2-EGFP plasmid successfully transfected Hep G2 hepatoma cells. The killing effect of HSV-tk suicide gene combined with cisplatin in Hep G2 cells was greater than using cisplatin or HSV-tk suicide gene alone(P0.05).Conclusion Our findings successfully assessed the killing effect of cisplatin combined with HSV-tk suicide gene in Hep G2 hepatoma cell line, providing evidence for clinical treatment for hepatoma.
