摘要 目的研究PAGE5基因在肝细胞癌(以下简称肝癌)样本及肝癌细胞株中的表达情况以及被沉默后对肝癌细胞株PLC/PRF/5及Focus细胞生长及转移能力的影响,分析PAGE5与甲胎蛋白(AFP)基因在肝癌组织样本中表达的相关性。方法采用半定量.PCR方法检测PAGE5在32例肝癌组织样本及其癌旁组织以及12株人肝癌细胞株中的表达;分析48例肝癌组织样本中PAGE5与AFP基因表达;同时观察干扰PAGE5表达后肝癌细胞增殖及转移能力的改变。结果在34.4%的肝癌组织样本中PAGE5基因表达上调;PAGE5基因表达与AFP表达水平呈正相关;利用RNA干扰技术沉默PAGE5基因表达后,PLC/PRF/5及Focus转移能力明显下降。结论 PAGE5基因在肝癌发生及维持肝癌细胞恶性表型中可能起到重要作用,深入研究PAGE5基因在肝癌中的功能,有望成为肝癌诊断标志物及潜在的治疗靶点。 Objective To study the expression profile of PAGE5 gene in HCC samples and cell lines, the corre- lation between PAGE5 and AFP in HCC samples and the effect on migration after PAGE5 RNAi in PLC/PRF/ 5 and focus cells. Methods Semi-quantitative RT-PCR was performed to detect the expression of PAGE5 in 32 samples of HCCs compared with adjacent non-cancerous livers and 12 HCC cell lines. The expressions of AFP gene were also evaluated in the 48 same HCC samples using RT-PCR. Cell progression and migration were evaluated after silencing of PAGE5 in Focus and PLC/PRF/5 cells. Results The expression of PAGE5 was significantly higher in HCC samples compared with the non-cancerous samples. The expression of PAGE5 and AFP correlated with each other well. Silencing of PAGE5 significantly promoted the migration of Focus and PLC/PRF/5 cells. Conclusion PAGE5 may play an important role in the hepatocarcinogenesis and may be used as a new marker or a potential therapeutic target for HCC.
