砷通过c-Jun氨基端激酶及蛋白激酶B信号传导通路对mdig基因表达的影响 被引量:2

摘要 mdig基因(mineraldust-inducedgene)亦称为mina-53及核蛋白52,2005年首先从煤矿工人的肺巨噬细胞中分离[1-3].在肺癌、乳腺癌、结肠癌、食道癌、胃癌、肝细胞癌、淋巴瘤及肾细胞癌等恶性肿瘤高表达,是一种潜在的新的致癌基因[4].砷(As)或砷化物是广为人知的致癌物[5],但砷致癌的机制尚不清.现有资料提示三氯化砷可通过激活促分裂原活化蛋白激酶(MAPK)[6]、转录激活因子AP-1[7]、p53[8]及核因子-κB[9]等信号通路而发挥致癌作用.二氧化硅粒子及烟草烟雾可诱导人支气管上皮细胞中mdig基因高表达,但砷化物与mdig基因表达的关系目前尚不清.本研究探讨三氯化砷与mdig基因表达的关系、职业性及环境性三氯化砷暴露致癌的发病机制.
