摘要
目的探讨心肌内注射成豪匆鰻胞生长因子(b FGF)和脑源性神摯营养因子(BDNF)缓释微粒对缺血心肌血管的生成作用。方法建立急性心肌梗死模型后,心肌缺血区分别注入生理盐水缓释微粒、b FGF缓释微粒及b FGF+BDNF缓释微粒。cine-MRI用于测量LVEF。免疫组织化学方法检测缺血区新生血管生成程度。结果 b FGF+BDNF组微血管密度比b FGF组和生理盐水组明显增高,b FGF+BDNF组LVEF明显增加。结论心肌内注射b FGF和BDNF缓释微粒可以促进缺血心肌的血管生成,提高左心室功能。b FGF和BDNF缓释微粒的联合应用可能是治疗缺血心肌的一种新的方法。
Objective The purpose of this study was to assess whether the simultaneous application of basic fibroblast growth factor (bFGF) and BDNF incorporating gelatin hydrogels improves angiogenesis and cardiac function in ischemic myocardium compared with bFGF applied alone. Methods Direct intramyocardial injection of 100 μg of bFGF plus 25 μg of BDNF microspheres, 100 μg of bFGF microspheres, or saline microspheres was performed in canine infarct model. Cardiac function was evaluated by cine-MRI. Myocardial microvessel density was assessed by von Willebrand factor immunohistochemical staining. Results LVEF was higher in bFGF plus BDNF group than in bFGF or saline group. The microvessel density in bFGF plus BDNF group were higher than that of bFGF or saline group. Conclusion This study indicates that the sustained dual release ofbFGF and BDNF incorporating gelatin hydrogels can improve angiogenesis and left ventricular function in the ischemic myocardium compared with bFGF applied alone, bFGF plus BDNF administration may be a promising therapeutic strategy for the treatment of ischemic myocardium.
