右归丸干预自身免疫性卵巢早衰小鼠EGF的实验研究 被引量:5

摘要 目的:观察右归丸对自身免疫性卵巢早衰小鼠模型血清FSH、E2、EGF及卵巢形态等的影响,以探讨右归丸的作用机理。方法:建立透明带抗原自身免疫性POF模型小鼠,并将其随机分为五组,空白组、模型组正常饮食、中药组(右归丸组)、西药组(补佳乐+黄体酮组)、中西组(右归丸+补佳乐+黄体酮组)。采用酶联免疫法检测血清中表皮细胞生长因子(Epidermal growth factor,EGF)水平,光镜下观察卵巢组织学形态,应用SPSS22.0进行统计分析。结果:造模后小鼠动情周期延长;光镜下观察卵巢组织,较空白组生长卵泡、黄体数少,闭锁卵泡多;血清中FSH、EGF值高于空白组(P<0.05),E2值低于空白组(P<0.05)。右归丸治疗后动情周期缩短;光镜下小鼠卵巢组织生长卵泡、黄体数较模型组增多,闭锁卵泡减少;血清FSH、EGF值降低(P<0.05),E2值升高(P<0.05)。中药组、西药组和中西组在对性激素及EGF水平的影响上无明显差异(P>0.05)。结论:1通过组织形态学、细胞生物学等研究证实右归丸能够从多方面促进小鼠卵泡生长、发育,改善卵巢功能;2右归丸能降低POF小鼠血清EGF水平;3EGF可有效反映自身免疫性POF模型小鼠卵巢功能的变化,可作为临床检测卵巢功能的指标。 Objective: To observe clinical effects of Yougui Wan on intervening serum FSH, E2, EGF and ovarian morphology in autoimmune premature ovarian failure mice model. Methods:POF model with autoimmune micezona pellucida antigen were randomly divided into five groups, as blank group, model group, TCM group (Yougui Wan group), western medicine group (Progynova+progesterone group), the integrative medicine group (Yougui Wan+Progynova+progesterone group). EGF levels in epidermal growth factor serum were observed by light microscope ovarian histology morphology. Results:After modeling, estrous cycle in mice was prolonged;ovarian tissue was observed by light microscope, growing follicles and corpus luteum was fewer than that in the blank, but the follicleatresia was in the contrary;FSH, EGF levels in serum was higher than that in the blank (P0.05). Conclusion: I. Depending on organizations morphology and cell biology research, Yougui Wan can promote the mice follicle growth, development and improve ovarian function;II. Yougui Wan can reduce EGF level in POF mice serum;III. EGF can effectively reflect the changes of ovarian function in autoimmune POF mice model, can be used as indicators in ovarian function detection.
