橙皮苷对四氯化碳致小鼠急性肝损伤的保护作用 被引量:2

摘要 目的探讨橙皮苷(HDN)预处理对四氯化碳(CCl4)所致小鼠急性肝损伤的保护作用及其机制。方法将30只雄性BALB/c小鼠随机分为正常组、CCl4组和HDN组。HDN组灌胃HDN(500 mg/kg),CCl4组和正常组灌胃同体积的0.5%羧甲基纤维素钠溶液,1次/d,连续14 d。末次灌胃后禁食不禁水,2 h后CCl4组和HDN组腹腔注射CCl4(0.3%,10 m L/kg)建立急性化学性肝损伤模型,正常组注射相同体积的橄榄油。16 h后按指标检测要求保留标本。测定各组小鼠肝、脾指数,肝组织匀浆丙二醛(MDA)、超氧化物歧化酶(SOD)和谷胱甘肽(GSH)的水平,血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)和高迁移率族蛋白B1(HMGB1)的水平;HE染色观察组织病理学改变;RT-PCR法检测小鼠肝组织HMGB1mRNA的表达水平。结果与正常组比较,CCl4组血清中ALT、AST水平显著升高(P<0.01),肝细胞水肿,脂肪变性,结构紊乱,肝细胞明显肿胀变形,有点状、片状坏死及炎性细胞浸润,MDA含量明显升高(P<0.01),SOD和GSH活性明显降低(P<0.01),血清中TNF-α和IL-6的水平显著升高(P<0.01),HMGB1的表达与释放明显增强(P<0.01)。与CCl4组比较,HDN组血清中ALT、AST水平显著降低(P<0.01),肝组织损伤程度明显减轻,MDA含量明显降低(P<0.01),SOD和GSH活性明显升高(P<0.01),TNF-α和IL-6水平显著降低(P<0.01),HMGB1的表达与释放受抑制(P<0.01,P<0.05)。结论 HDN对CCl4所致小鼠急性肝损伤具有保护作用,其机制可能与抑制脂质过氧化,减少TNF-α及IL-6的产生以及抑制HMGB1的表达和释放有关。
