Determination of 6-Hydroxykynurenic Acid in Ginkgo Preparation by HPLC

摘要 目的建立银杏叶制剂中6-羟基犬尿喹啉酸(6-HKA)含量的测定方法。方法色谱柱为XDB-C8(250 mm×4.6 mm,5μm);流动相为0.1%磷酸-乙腈-甲醇(94∶5∶1);流速1.0 mL·min^-1;检测波长为350 nm。制剂样品经粉碎后,用50%甲醇溶解,过滤后进样测定。结果 6-HKA在5.025-100.5μg·mL^-1内线性良好,峰面积与进样量呈良好线性关系,r=1.000 0;平均回收率为99.86%。6批银杏叶片剂中6-HKA的含量为0.22%-0.35%。结论建立的方法操作简便,结果准确,重复性好。 OBJECTIVE To establish the method of determining the 6-hydroxykynurenic acid(6-HKA) content with HPLC. METHODS XDB-C8 column(4.6 mm×250 mm, 5 μm) was used. Mobile phase was mixture of 0.1% phosphoric acid-CH3CN-CH3OH(94∶5∶1). The flow rate was 1.0 mL·min^-1. The UV detector was operated at 350 nm. Tablets were comminution, dissolution with 50% methanol, and determination after filtration. RESULTS The linear range of 6-HKA was 5.025-100.5μg·mL-1(r=1.000 0); the average recovery was 99.86%. The content of 6-HKA in 6 batches of Ginkgo tablets was 0.22%-0.35%. CONCLUSION The method is simple, the result is accurate, and the reproducibility is good.
