摘要
目的探讨丹左合方对AD模型大鼠血清CRP、COX-2、IL-6含量,脑海马组织NF-κB、iNOS阳性表达的影响。方法 SD大鼠40只,随机分为正常组、丹左合方组、对照组、模型组,每组10只。采用D-半乳糖颈背部皮下注射6周制备AD大鼠模型。各手术组大鼠经相应药物灌胃治疗5周后,ELISA法检测大鼠血清CRP、COX-2含量,放免法检测血清IL-6含量,免疫组化法检测大鼠脑海马组织NF-κB、iNOS的阳性表达。结果 CRP、COX-2、IL-6含量丹左合方组显著降低,与模型组比较有显著性差异(P<0.01),与对照组比较有显著性差异(P<0.01);丹左合方组、对照组、模型组海马组织胞核内可见棕黄色的NF-κB颗粒,胞浆内可见棕黄色的iNOS颗粒,模型组的棕黄色颗粒最多,表明NF-κB、iNOS阳性表达最强,丹左合方组棕黄色颗粒明显少于模型组、对照组;丹左合方组NF-κB、iNOS阳性细胞数量与模型组比较有显著性差异(P<0.01),与对照组有显著性差异(P<0.01)。结论丹左合方可降低AD大鼠血清CRP、COX-2、IL-6的含量,降低AD大鼠海马NF-κB、iNOS的阳性表达,起到减缓AD大鼠炎性反应的作用。
Objective To investigate the expression effects to NF-κB,iNOS in hippocampus and CRP,COX-2,IL-6 in serum of rats with Alzheimer disease. Methods 40 SD rats were randomly divided into normal group,control group,model group,Dan Zuo Hefang group,with 10 rats in each group. The rat model of AD was made by D-galactose subcutaneous injection through 6weeks. The operation group rats were gavaged administration with different drugs through 5 weeks,after 5 weeks of gavage,serum CRP,COX-2 content with ELISA method,the samples were determined by equilibrium method,according to RIA kit instructions steps were measured IL-6,immunohistochemistry was used to detect the positive expression of NF-κ B,iNOS in hippocampus with rat brain. Results The content of CRP,COX-2,IL-6 in Dan Zuo Hefang group decreased significantly,there were significant differences compared with the model group(P〈0. 01),there was significant difference compared with the control group(P〈0. 01); control group iNOS NF-κ B,NF-κ B expression was negative,particle Dan Zuo Hefang group,control group,model group,hippocampal tissue nucleus can be seen in the brown color,iNOS particles in the cytoplasm of brown yellow,brown yellow granules in model group showed up,NF-κ B,iNOS positive expression was strongest; Dan Zuo Hefang group of NF-κB,the positive expression of iNOS was decreased than that in model group(P〈0. 01),significantly reduced compared with the control group(P〈0. 01). Conclusion Dan Zuo Hefang can reduce the content of serum CRP,COX-2,IL-6 of AD rats,decrease the positive expression of NF-κ B,iNOS in hippocampal with rat AD,to slow down the inflammatory response in AD rats.
