艾片与合成冰片对脑缺血再灌注损伤大鼠脑保护的比较研究 被引量:11

Comparison of the brain-protect effects on middle cerebral artery occlusion rats model and factors influencing between L-borneolum and borneolum syntheticum

摘要 目的比较艾片与合成冰片对大鼠脑缺血再灌注模型(MCAO)的脑保护及其影响机制。方法分别给予大鼠艾片200mg/kg、133.33mg/kg及合成冰片200mg/kg、133.33mg/kg,参照Zea Longa法,对大鼠施与MCAO手术,于再灌注4h、22h测量体温,并检测灌注22h后大鼠脑水肿率以及脑组织钙离子含量。结果与模型组比较,灌注4h时艾片200mg/kg组能显著降低模型大鼠体温(P<0.05);艾片133.33mg/kg、合成冰片200mg/kg组有抑制大鼠体温的趋势,但无显著性差异(P>0.05);艾片与合成冰片的200mg/kg组,均能显著降低灌注22h的模型大鼠体温(P<0.01,P<0.05)。艾片与合成冰片的200,133.33 mg/kg组均能显著降低模型大鼠脑水肿率(P<0.01,P<0.05);并能极显著降低缺血侧脑组织中Ca2+含量(P<0.01)。结论艾片与冰片均能改善脑缺血大鼠模型的体温升高,并可降低脑水肿和钙离子而发挥脑保护作用,初步表达出冰片的寒凉药性与清热开窍的"性-效"关联特征,且同剂量艾片有优于合成冰片的趋势,仅从药效角度考虑,急救用药选择艾片较理想。 Objective To compare the brain-protect effects on middle cerebral artery occlusion( MCAO) rats model and factors influencing between L-borneolum and borneolum syntheticum. Methods After according model big rats L-borneolum( 200 mg /kg,133. 33 mg /kg) and borneolum syntheticum( 200 mg /kg,133. 33 mg /kg) separately,do MCAO operation in Zea Longa method on big rats. Test body temperature 4h,22 h after reperfusion,and detect big rats' brain edema rate and content of calcium ion in brain tissue. Results 1Compared with model group,L-borneolum( 200 mg /kg) can apparently bring down body temperature of model big rats(P〈0. 05). 4h after reperfusion. Within L-borneolum( 133. 33 mg /kg) and borneolum syntheticum( 200mg/kg) exists trend of body temperature inhibition,but no apparent difference(P〈0. 05). Both L-borneolum( 200mg/kg) and borneolum syntheticum( 200 mg /kg) show apparent decline of big rats' body temperature after 22h(P〈0. 01,P 0. 05),2Both L-borneolum( 200,133. 33 mg/kg) and borneolum syntheticum( 200,133. 33 mg/kg) can apparently bring down model big rats' brain edema rate(P〈0. 01,P 0. 05),and extremely apparently bring down content of calcium ion in ischemia side of brain tissue. Conclusion L-borneolum and borneolum syntheticum can both show positive effects on lift of body temperature caused by MCAO,while creating brain-protection effects by bringing down brain edema rate and content of brain tissue calcium ion,which primarily reflects borneolum syntheticum's characteristic of 'property-effect' connection between its cold property and heat-clearing and orifice-opening effects. There exists a trend that L-borneolum comes better than borneolum syntheticum in equal dose. So considering in aspect of effect only,it is more ideal to choose borneolum syntheticum as first aid medicine.
