摘要
目的观察赤雹根总皂苷(TSTR)对2型糖尿病(T2DM)模型大鼠血糖、血清胰岛素(INS)、胰岛素抵抗指数(IRI)、胰岛素敏感指数(ISI)及糖化血红蛋白水平(GHb)的影响,探讨TSTR对T2DM的治疗作用。方法 Wistar大鼠,采用高糖高脂饲料加链脲佐菌素(STZ)制备T2DM模型,3周血糖>16.7 mmol/L为造模成功,将模型大鼠随机分为模型对照组、TSTR大剂量组(160 mg·kg-1·d-1)、TSTR小剂量组(80 mg·kg-1·d-1)、盐酸二甲双胍片(MHT)100 mg·kg-1·d-1阳性对照组,各给药组分别ig相应药物,模型组ig等体积的蒸馏水。测定首次给药后6 h内血糖及连续给药第3周血糖、INS、及GHb水平并计算IRI和ISI指数。结果首次给药后TSTR大、小剂量组4.6时间点血糖显著低于模型组(P<0.05或P<0.01);连续给药3周,TSTR大、小剂量组血糖、INS、IRI和GHb均显著低于模型对照组(P<0.05或P<0.01);ISI均显著高于模型组(P<0.01)。结论 TSTR对大鼠T2DM有治疗作用,其机制可能与其降低IRI,升高ISI有关。
Objective To investigate the therapeutic effect of TSTR in T2 DM by observing effect of total saponin from Thladiantha dubia roots( TSTR) on blood glucose,insulin( INS),insulin resistance index( IRI),insulin sensitivity index( ISI),rats were prepared with high sugar,high fat diet and streptozotocin( STZ),whose glycated hemoglobin( GHb) in rats with T2 DM. Methods T2 DM model of Wistar blood glucose were 16. 7mmol /L after 3 weeks. T2 DM model rats were randomly divided into model group,positive control group which were ig with metformin hydrochloride tablets( MHT,100 mg·kg-1·d-1),big dosage group of TSTR( 160 mg·kg-1·d-1) and small dosage group of TSTR( 80 mg·kg-1·d-1). Each group ig corresponding drugs respectively,and model group ig same volume of distilled water. The level of blood glucose of T2 DM rats were determined within 6hours after first ig TSTR and the level of blood glucose,INS and GHb of T2 DM rats were determined in1 st,2nd and 3rd weeks after administration. Results 2nd,4th and 6th hours after administration,the blood glucoses of all the TSTR dose groups were significantly lower than those of model group(P〈0. 05 orP〈0. 01). After 3 weeks administration,the blood glucoses content,INS,IRI and GHb were significantly lower than those in model group(P〈0. 01) and ISI were significantly higher than those in model group(P〈0. 01) in big and small dose groups of TSTR. Conclusion TSTR has a therapeutic effect on T2 DM in rats,its mechanism of action may be associated with decreased IRI,increased ISI.
