VDR基因多态性与宁夏回族儿童哮喘相关性研究 被引量:3

摘要 目的探讨VDR(维生素D受体)基因BsmI、ApaI、TaqI和FokI位点的单核苷酸多态性与宁夏回族哮喘发病的关系。方法采用PCR-RFLP方法分析VDR基因4个多态位点(FokI,BsmI,TaqI,APaI)的基因型及基因频率。结果 FokI位点的基因型分布与基因频率分布在宁夏回族病例组与宁夏回族对照组及在回族病例组与汉族病例组之间差异有统计学意义(P<0.05),表明FokI位点可能是宁夏回族儿童的一个哮喘易感位点,同时也可能是宁夏回族儿童哮喘与宁夏汉族儿童哮喘之间存在差异性的一个位点。BsmI、ApaI及TaqI位点的基因频率与基因型频率分布在宁夏回族病例组与宁夏回族对照组及在宁夏回族病例组及宁夏汉族病例组之差异无统计学意义(P>0.05)。结论 VDR基因的FokI位点与宁夏回族儿童哮喘存在显著相关性,可能是宁夏回族儿童哮喘的易感因素。 Objective To discuss the relationship between the single nucleotide polymorphisms of locus of BsmI, ApaI,TaqI and FokI on the VDR gene and Ningxia Hui children with asthma and the differences between Ningxia Hui and Han asthmatic children. Methods The peripheral blood DNA was extracted from Hui and Han asthmatic children and from normal Hui group, and the genotype and gene frequency of dynamic locus of VDR gene were analyzed by using PCR and PCR - RFLP. Results The distinguished differences about the distribution of genotype and gene frequency of FokI locus existed between the Ningxia Hui Asthma group and Ningxia Hui Nor- mal group and Hui asthma group and Han Asthma group. The results showed that FokI locus was an asthma susceptibility locus and it was a locus where the difference of asthma existed between Ningxia Hui Children and Ningxia Han children. There was no distinguished differ- ence about the distribution of gene frequency and genotype frequency of the locus of BsmI, ApaI and TaqI existed between the Ningxia Hui asthma group and Ningxia Hui normal group and Hui asthma group and Han asthma group. Conclusion There is a significant corre- lation between FokI locus and Ningxia Hui children's asthma: Fokl locus is a locus where the differences existed between Ningxia Hui children's asthma and Ningxia Han children's asthma. There are some differences among the asthma susceptibility loci of candidate VDR gene of asthma gained by different ethnic groups.
