miR-106b在原发性肝癌中的表达及与预后的关系 被引量:4

摘要 目的探讨miR-106b在肝癌(HCC)组织中的表达及其临床意义。方法选择10对HCC组织及相应正常肝组织标本及82例有随访资料的HCC组织切片。qRT-PCR法及锁定核酸原位杂交检测miR-106b在HCC组织的表达,分析miR-106b表达与患者临床病理特征及预后的关系。结果 miR-106b在HCC组织中表达上调(P<0.05)。miR-106b表达与转移、组织分级、肝硬化及血清甲胎蛋白明显相关(P=0.046、0.019、0.004及0.043),而与年龄、性别、乙型肝炎、肿瘤大小及门脉癌栓无明显相关。HCC组织miR-106b高表达的患者5年生存率明显低于低表达患者(P<0.01),miR-106b高表达是影响HCC患者总体生存的独立风险因素(P<0.05)。结论 miR-106b在HCC组织中表达明显上调,可能参与了HCC的发展过程,有望成为一种新的HCC预后参考指标。 Objective To detect the expression of miR-106 b in hepatocellular carcinoma(HCC),and to investigate its correlaion with clinicopathologic features. Methods The qRT- PCR analysis and locked nucleic acids- in situ hybridization analysis were used to assess the expression of miR- 106 b in 10 paired HCC tissues and 82 HCC tissues with completed clinic followed- up data. Statistical analysis was used to determine the correlation of miR- 106 b expression with the clinicopathologic features and prognosis of patients. Results Significant up- regualtion in miR- 106 b was observed in HCC tissues(P〈0. 05). High miR- 106 b expression was significantly correlated with the metastasis,degree of differentiation,cirrhosis and serum AFP(P〈0. 05); but not with age,gender,hepatitis B,tumor size or portal vein embolism. Patients with high miR- 106 b expression had significantly poorer overall survival(P〈0. 01). High miR- 106 b expression was independent prognostic factors for overall survival(P〈0. 05). Conclusion The expression of miR- 106 b is highly up- regulated in HCC tissues,indicating that high miR- 106 b expression may be involved in the process of metastasis and suggesting a novel prognosis reference of HCC.
