HIF-1α、VEGF-A和MMP-9在脊索样脑膜瘤中的表达及意义 被引量:1

摘要 目的研究缺氧诱导因子1α(hypoxia-inducible factor 1-alpha,HIF-1α)、血管内皮生长因子A(vascular endothelial growth factor a,VEGF-A)、基质金属蛋白酶9(matrix metalloproteinase 9,MMP-9)与脊索样脑膜瘤(chordoid meningioma,CM)血管新生和浸润的关系。方法采用免疫组化、蛋白印迹分析(Western blot)和酶联免疫法(RT-PCR)检测HIF-1α、VEGF-A、MMP-9在蛋白质和mRNA两个水平上的表达,同时采用CD34标记微血管密度(microvessel density,MVD),研究HIF-1α、VEGF-A、MMP-9与CM血管新生及复发的相关性。结果与正常脑组织和WHOⅠ级脑膜瘤相比,HIF-1α、VEGF-A、MMP-9在CM中明显表达(P<0.05),与非典型脑膜瘤相比,CM中表达虽多但差异无统计学意义(P>0.05);HIF-1α、VEGF-A在与MVD呈正相关且差异有统计学意义(r1=0.930,r2=0.916,均P<0.01)。结论 HIF-1α、VEGF-A、MMP-9促进CM新血管生成,进一步影响肿瘤浸润和复发,可做为术后辅助抗血管治疗以降低其复发率的分子靶点。
