摘要
目的探讨促炎因子与抑炎因子的失衡在多囊卵巢综合征(PCOS)中的重要作用以及吡格列酮、二甲双胍在维持两者之间平衡的作用。方法将50只雌性SD大鼠随机分为5组,其中40只建立PCOS雌鼠模型后,分别予生理盐水、二甲双胍、吡格列酮以及后面两者联合干预,而10只成年正常雌性SD大鼠,仅予生理盐水灌胃干预。取干预前后雌鼠的血浆行固相夹心法酶联免疫吸附实验(ELISA),检测血清超敏C反应蛋白(hs-CRP)、降钙素原(PCT)、白细胞介素-10(IL-10)与脂联素(ADP)含量变化。结果 PCOS雌鼠hs-CRP、PCT水平较正常雌鼠升高,ADP水平较正常雌鼠下降,差异均有统计学意义(P<0.01);IL-10水平较正常雌鼠下降,差异有统计学意义(P<0.05)。PCOS雌鼠经不同方法干预后,尤其在吡格列酮联合二甲双胍干预雌鼠,hs-CRP、PCT水平较干预前降低,IL-10、ADP水平较干预前升高,差异均有统计学意义(P<0.01)。结论 PCOS雌鼠促炎因子与抑炎因子处于失衡状态,二甲双胍、吡格列酮联合干预可能纠正这种慢性低度炎症状态。
Objective To investigate the role of imbalance of proinflammatory and inflammation inhibitory cytokine in polycystic ovary syndrome(PCOS) and the effects of pioglitazone and metformin on this process. Methods Fifty female rats were randomly divided into 5 groups(n = 10). PCOS models were established in 4 groups,and treat with normal saline,metformin,pioglitazone or metformin plus pioglitazone. Yet the 10 normal matured female rats were fed only with normal saline as controls. ELISA was applied for assessment of serum hs- CRP,PCT,IL- 10 and ADP before and after treatment. Results Significant elevations of serum hs- CRP and PCT with reductions in ADP and IL- 10 were observed in rats with PCOS than controls. Significant improvements in hs- CRP,PCT,IL- 10 and ADP were observed in all treatment groups,especially in combined treatment group,after treatment( P〈0. 01). Conclusion The proinflammatory and inflammation inhibitory cytokines in rats suffering from PCOS were in unbalanced state. Metformin and pioglitazone combined treatment may remedy this state of chronic low- grade inflammation.
