骨髓间充质干细胞对脑缺血新生小鼠运动行为状态异常的疗效观察

摘要 目的观察人骨髓间充质干细胞对脑缺血小鼠行为的影响。方法 40只7~8日龄新生鼠随机分为假手术组13只,仅做颈部皮肤的切开缝合,未结扎双侧颈总动脉;手术组12只,结扎双侧颈总动脉;干细胞移植组15只,结扎双侧颈总动脉后4 d,给本组小鼠心腔注射骨髓间充质干细胞0.15 ml(含骨髓间充质干细胞106)。同时给假手术组和手术组小鼠心腔注射0.15 ml生理盐水。应用动物行为学平台(转棒仪和旷场试验)对36~37日龄小鼠进行运动和行为功能测定。运动状态以及行为状态假手术组和手术组以及干细胞移植组3组采用随机F检验及两两比较。结果 (1)干细胞移植组和假手术组小鼠运动降落的潜伏期明显长于手术组,而假手术组和干细胞移植组的小鼠运动降落的潜伏期长短无差异(F=8.525,P=0.001);(2)在旷场实验中3组小鼠跨格次数,运动的总路程以及平均速度比较均无差异(均P〉0.05);(3)干细胞移植组和假手术组小鼠周边活动时间和距离明显长于手术组(F=8.370,P=0.001),而假手术组和干细胞移植组的小鼠周边活动时间和距离长短无差异(F=3.541,P=0.039);(4)干细胞移植组和假手术组小鼠中心活动时间及路程明显短于手术组(F=7.201,P=0.002),干细胞移植组和假手术组小鼠中心活动的时间和距离比较无差异(F=6.044,P=0.005)。结论脑缺血小鼠的运动和行为状态明显受损,骨髓间充质干细胞移植可明显改善脑缺血小鼠运动和协调能力和认知能力。 Objective To examine the effects of human bone marrow mesenchymal stem cells on behaviors of cerebral ischemia mice. Methods 40 neonatal mice of 7-8 day old were randomly divided into sham group with only neck skin incision and no carotid artery ligation, surgery group with the ligation of bilateral common carotid artery and stem cell transplantation group with the ligation of bilateral common carotid artery. After 4 days, neonatal mice in the stem cell transplantation group were injected with 0.15 ml of bone marrow mesenchymal stem cells(1×10^6 cells) to the heart of neonatal mice. Neonatal mice in the sham group and surgery group received only an injection of 0.15 ml saline. After 30 days mice were evaluated for the movement and behavioral functions with the ethology platform(bar-transferring meter and open-field test). Results(1) The time at which each animal fell from the drum was much longer in the stem cell transplantation group and the sham group than those in the surgery group, and was not significantly different between the sham group and stem cell transplantation group(F = 8.525,P = 0.001).(2) In the open field test, the grid-crossing frequency, the total distance and the average movement speed of mice between three groups were not significantly different(P〈0.05).(3) The time and distance of peripheral activity of mice in the stem cell transplantation group and the sham group were much longer than those of mice in the surgery group(F = 8.370,P = 0.001), and there was difference between the sham group and the stem cell transplantation group(F = 3.541,P = 0.039).(4) The time and distance of center activity of mice in the stem cell transplantation group and the sham group were much shorter than those of mice in the surgery group(F = 7.201,P = 0.002), and there was no difference between the sham group and the stem cell transplantation group(F = 6.044,P = 0.005). Conclusion The transplantation of bone marrow mesenchymal stem cells can significantly improve the movement, coordination skills and cognitive abilities of cerebral ischemia mice.
