参附汤入血有效成分对急性心衰大鼠血流动力学及心肌凋亡、自噬的影响 被引量:9

摘要 目的:研究参附汤入血有效成分对急性心衰大鼠血流动力学及心肌凋亡、自噬的影响,以探讨其回阳救逆的作用机制。方法:①采用盐酸异丙肾上腺素腹腔注射建立大鼠急性心衰动物模型,用16导生理记录仪同步记录左室内压(LVSP)、左室内压最大上升速率(+dp/dtmax)、左室内压最大下降速率(-dp/dtmin),动态观察参附汤入血有效成分对急性心衰大鼠血流动力学的影响。②采用实时荧光定量PCR,以SYBRGreen I法测定自噬相关基因Beclin-1,Atg5mRNA表达量、凋亡相关基因Caspase mRNA的表达量。结果:①模型组与空白组比较,左心室内压、左心室内压最大上升速率及左心室内压最大下降速率有明显降低,说明模型成立;参附汤水煎液组、参附汤萃取液组、阳性组与模型组比较,左心室内压、左心室内压最大上升速率及左心室内压最大下降速率比较差异有统计学意义(P<0.01);②与模型组比较,参附汤水煎液组、参附汤萃取液组与阳性对照组Beclin1、Atg5mRNA的表达量均明显下降,与模型组比较,参附汤水煎液组、参附汤萃取液组与阳性对照组Caspase mRNA的表达量均明显下降。结论:参附汤入血有效成分对急性心衰大鼠血流动力学指标有明显的改善作用,且参附汤水煎液与参附汤萃取液给药后改善作用差异无统计学意义;参附汤入血有效成分可保护急性心衰大鼠心肌细胞,有抑制细胞凋亡和自噬的作用,且与降低Beclin-1、Atg5 mRNA、Caspase mRNA的表达量有关。 Objective : Study on effect of Shenfu Decoction into blood ingredients on acute heart failure hemodynamics and myocardial apoptosis, autophagy in rats, and to investigate the mechanism of action of reviving yang. Method:(1)The acute heart failure rat animal model was established by intraperitoneal injection of isoprenaline hydrochloride, 16 polygraph record left ventricular pressure (LVSP) , LVP ( + dp/dtmax) , The largest decline rate of left ventricular pressure (-dp/ dtmin), Dynamic observation of Shenfu Decoction into blood ingredients on hemodynamics in rats with acute heart failure effect(2)By real-time fluorescent quantitative PCR Mx3000P, autophagy related gene Beclin-1 was measured with SYBR Green I, expression of Atg5 mRNA, the expression of apoptosis related gene Caspase mRNA. Result:(1)The model group compared with the control group the internal pressure of left ventricle, left ventricular pressure and maximum rate of rise of left ventricular pressure maximum falling rate has decreased, the model was established; Shenfu Decoction group, Shenfu Decoction extract group, positive group compared with the model group, the internal pressure of left ventricle, left ventricle the maximum rate of rise of pressure and left ventricular pressure maximum falling rate had significant differences ( P 〈 0.01 ). (~)Compared with the model group, Shenfu Decoction group, the expression of Shenfu Decoction extract group and positive control group Beclinl, Atg5 and mRNA decreased obviously. Compared with the model group, Shenfu Decoction group, the expression of Shenfu Decoction extract group and positive control group Caspase, mRNA decreased obviously. Conclusion : Shenfu Decoction into blood components is effective in the treatment of acute heart failure in rat blood dynamics index. Shenfu Decoction into blood components could protect myocardial cells in rats with acute heart failure, according to inhibition of apoptosis and autophagy, and decreased Beclin-1, Atg5 mRNA, Caspase mRNA expression.
