福州市乙型流感病毒HA1基因特性的分析

摘要 目的了解福州市2006年-2012年流行的乙型流感病毒血凝素基因变异特点及种系进化分布。方法采用狗肾传代细胞培养分离流感病毒,选取部分乙型流感病毒株进行血凝素基因HA1区核苷酸序列测定并推导出其氨基酸序列,然后用DNASTAR MegAlign序列分析软件进行基因种系进化特性分析。结果 2006年-2012年福州市人群中同时流行着乙型Yamagata系和Victoria系毒株。分离的Yamagata系毒株与B/Florida/4/2006比较,在血凝素基因HA1区发生3个~7个氨基酸替换。分离的Victoria系毒株与B/Brisbane/60/2008比较,在HA1区发生5个~7个氨基酸替换,在197位增加一个糖基化位点。结论福州市人群中流行的乙型流感病毒与Yamagata系和Victoria系代表株相比基因特性已经发生了进一步改变。 Objective To understand the variation and epidemic characteristics of influenza B virus from 2006 to 2012 in Fuzhou city through hemagglutinin gene detection.Methods Influenza B virus were isolated with MDCK cells.Then some influenza B virus strains were selected to determine the nucleotide sequences of HA1 gene and deduce the corresponding amino acid sequences.The phylogenetic tree was analyzed with DNASTAR MegAlign soft ware.Results The Yamagata-like and Victoria-like viruses co-circulated in Fuzhou from 2006 to 2012.Compared with B/Florida/4/2006,there were 3 ~ 7 amino acids substitution in HA1 region of Yamagata-like isolates.Compared with B/Brisbane/60/2008,the Victoria-like isolates were found 5 ~ 7 amino acids mutation in the HA1 region,and a more glycoaylation site at 197thsite.Conclusion The genetic characteristics of prevailed influenza B viruses in Fuzhou has changed in comparing with the Yamagata-like and Victoria-like representative strains.
