微量RhD抗原检测法

Trace RhD antigen detection method

目的为解决患儿、烧伤患者以及危重患者,因采集RhD抗原检测的血标本困难,而研究的一种微量RhD抗原检测方法。方法取洁净玻片一张,用加样器加入RhD单克隆试剂血清和被检测患者5%红细胞悬液各10μl,再加入低离子介质溶液20μl于玻片上。将上述3种液体混匀,置室温5 min观察结果。对幼儿园206份儿童体检血标本与常规玻片法进行检测比较。结果 206份儿童血标本微量法与常规法平行对照,2种方法检测RhD抗原结果完全吻合。结论该方法操作简便、价格低廉,且血标本和试剂用量少,适用于儿童、大面积烧伤患者以及危重患者,进行RhD抗原检测。

Objective To study a trace RhD antigen detection method,so as to solve the difficulty in sampling blood specimens from children,burn patients and critical patients for RhD antigen detection in.Methods Both RhD monoclonal serum reagent and 5% suspension of red blood cells from patients at a volume of 10 μl,and 20 μl low ionic medium solution were put onto a clean slide by pipette,then mixed evenly and kept for 5 min at room temperature to observe the change.Blood specimens of 206children were collected to detect RhD antigen by micro method and conventional slide method respectively,and the results were compared.Results RhD antigen detection results of the two methods were the same by parallel-control trial of the 206 children blood samples.Conclusion This method has the advantages of simple operation,low price,less-dosage reagents and blood samples,so it is suitable for RhD antigen detection in children,patients with large-area burn and critical patients.
