摘要
目的:采用1H核磁共振(1H NMR)代谢组学技术研究胆固醇胆石豚鼠血清的特征性代谢物。方法:30只健康成年雄性豚鼠随机分为模型组和对照组,模型组和对照组分别服用致石饲料和常规饲料各8周。第9周第一天清晨空腹采集所有豚鼠腹腔动脉血1m L,采用1H NMR技术检测血清小分子代谢物;摘取胆囊收集胆汁,观察胆石形成情况。结果:模型组胆固醇胆石成石度明显高于对照组;与对照组相比,模型组血清丙氨酸、柠檬酸、丙酮、谷氨酸、甘氨酸、赖氨酸、蛋氨酸、苯丙氨酸和酪氨酸等含量明显升高,乳酸、丙酮酸和谷氨酰胺明显降低,说明模型组存在严重的能量、脂质及蛋白质代谢紊乱。结论:豚鼠胆固醇胆石的形成与能量、脂质及蛋白质代谢紊乱有关。
Objective: To explore the serum characteristic metabolites in guinea pigs with cholesterol cholecystolithiasis by using1 H NMR- based metabonomics. Methods: 30 healthy adult male guinea pigs were allocated to model group and control group,each group contained 15 guinea pigs. Guinea pigs in model group and control group were given lithiasis- producing fodder and routine fodder respectively for 8 weeks. On the first day's morning of the 9thweek,1m L of fasting blood from coeliac artery of each guinea pig in two groups were taken for measurement and analysis of small molecular metabolites in serum by using1 H NMR- based metabonomics technique,and the gallbladders were resected and the biles were collected to observe the cholesterol cholecystolithiasis formation. Results: The cholesterol cholecystolithiasis- producing degree in model group guinea pigs were significantly higher than that in control group guinea pigs. The serum levels of alanine, citrate, acetone, glutamate, glycine, lysine,methionine,phenylalanine and tyrosine significantly increased,while lactate,pyruvate and glutamine levels decreased significantly in the model group guinea pigs,to compare with those in control group,obvious disturbance of energey,lipid and protein metabolism were existed in model group. Conclusion:The formation of cholesterol cholecystolithiasis in guinea pigs is associated with obvious disturbance of energy,lipid and protein metabolism.
