小菜蛾抗菌肽的纯化工艺研究

摘要 目的:优化小菜蛾抗菌肽(CA)的纯化工艺。方法:以总多肽含量、CA含量及Hep G2细胞抑制率为评价指标,选择溶剂类型、提取次数和溶剂用量为考察因素,采用L9(34)正交试验法优选小菜蛾抗菌肽最佳纯化工艺。结果:最优的纯化工艺条件为A3B3C2,即采用p H 6.5磷酸盐缓冲液纯化3次,每次5倍量。结论:该工艺可用于小菜蛾抗菌肽的纯化,其工艺稳定可行,为其新药产业化提供参考。 Objective: To optimize the purification process of antimicrobial peptides of P xylostella. Methods: Studying the content of total peptide, the content of CA and the suppression ratio of Hep G2 cells as the main indicators, solvent type, extraction times and the amount of solvent as factors. Using L9 (3^4) orthogonal experiment method to determine the optimum purification of antimicrobial peptides P xylostella. Results: The optimum purification process was A3B3C2, with pH 6.5 phosphate buffer purification three times, each time with 5-fold amount of the solvent. Conclusion: The process can be used for the purification of antimicrobial peptides of diamondback moth, which is stable and feasible, and to provide a reference for their new drug industriatization.
