摘要
目的通过检测人类免疫缺陷病毒(HIV)窗口期标本,比较目前中国使用的几种HIV酶联免疫吸附分析(ELISA)检测试剂的灵敏度。方法 1例HIV核酸检测阳性、酶联免疫阴性首次献血者标本,于第1、2、3周各抽取血3份。用检测方法A(中国产第4代HIV ELISA检测试剂A)、检测方法 B(中国产第3代HIV ELISA检测试剂B)、检测方法 C(进口第4代HIV ELISA检测试剂C)分别进行常规抗体检测,并用生物素-亲和素ELISA对该血样本的HIV-1 P24抗原进行了单独检测。结果用检测方法 B检测时,第1、2、3周标本均显示为阴性;用检测方法 A检测时,第1、2周标本均显示为阴性,第3周标本显示为阳性;用检测方法 C检测时,第1周标本显示为阴性,第2、3周标本均显示为阳性。用生物素-亲和素ELISA检测HIV-1 P24抗原,结果显示第1、2周标本HIV-1 P24抗原均显示为阴性,第3周标本HIV-1 P24抗原显示为阳性。结论实验所用检测方法 A的中国产第4代HIV检测试剂A的灵敏度比检测方法 B的中国产第3代HIV检测试剂B灵敏度高,但比实验所用检测方法 C的进口第4代HIV检测试剂C灵敏度低。
Objective To detect human immunodeficiency virus (HI.V) window period specimens, and compare the detection sensitivity for current domestic detection agents of HIV enzyme linked immunosorbent assay (ELISA). Methods One first blood specimens of donor with positive HIV nucleic acid testing and negative enzyme -linked immunosorbent was used to depict 3 specimens at 1-week, 2-week, and 3-week. Three methods were performed respectively in routine antibody detection: method A (domestic 4th generation HIV ELISA test kit A), method B(domestic 3rd generation HIV ELISA test kit B), and method C(imported 4th generation HIV ELISA test kit C), and the blood samples of HIV-1 P24 antigen were detected separately by biotin-streptavidin- biotin ELISA. Results The specimens showed as negative at 1-week, 2-week and 3-week in method A; the specimens showed as negative at 1-week and 2-week, and specimens at 3-week showed as positive in method B; the specimens showed as negative at 1-week, and specimens at 2-week and 3-week showed as positive in method C. The HIV-1 P24 antigen were detected by biotin-avidin-biotin ELISA, the results of HIV-1 P24 antigen specimens showed as negative at 1-week and 2-week and showed as positive at 3-week. Conclusion It is demonstrated that the sensitivity of detection reagent in method A is higher than that of method B, but lower than method C.
