摘要
目的探讨miRNA-449a在肺癌细胞株中的表达,及其对肺癌分化程度的影响。方法应用荧光定量PCR检测法对40例肺癌患者(肺癌组)及20例非肺癌患者(非肺癌组)肺组织活检细胞中的miRNA-449a进行检测,比较其在肺癌组与非肺癌组中的检出差异。同时根据肺癌分化程度将肺癌组分为高分化组(16例)与低分化组(24例),比较miRNA-449a在不同分化组织中的检出差异。结果肺癌组miRNA-449a的阳性检测率明显低于非肺癌组(P<0.05),肺癌高分化组miRNA-449a的阳性检测率显著高于肺癌低分化组(P<0.05);此外,肺癌组95D-p CDNA3.1-miR-449a划痕宽度明显低于非肺癌组(P<0.05),肺癌高分化组95D-p CDNA3.1-miR-449a划痕宽度显著高于肺癌低分化组(P<0.05)。结论 miRNA-449a可作为筛查肺癌及评估肺癌分期的一项重要指标。
Objective To explore the expression of miRNA-449 a in lung cancer cell line,and its effect on degree of differentiation of lung cancer cells.Methods Fluorescent quantitative PCR assay had been applied for examination of micrornas 449 a in cells of lung biopsies of 40 patients with lung cancer( lung cancer group) and 20 patients with non-lung cancer( non lung cancer group),and results of detection had been compared between lung cancer group and non lung cancer group.At the same time,patients in lung cancer group were further divided into high differentiation group( 16 cases) and low differentiation group( 24 cases) according to the degree of differentiation of lung cancer cells and the difference in expression of miRNA-449 a in tissue with different degree of differentiation was compared between these two groups.Results The positive detection rate of miRNA-449 a in non lung cancer group was significantly lower than lung cancer group( P0.05).The positive detection rate of micrornas-449 a in well differentiated lung cancer group was significantly higher than that of low differentiated group( P0.05).In addition,the scratch width of 95 d-p CDNA3.1-miR-449 a in lung cancer group was significantly lower than that of non lung cancer group( P 0.05),and the scratch width of 95-d-p CDNA3.1 miR-449 a in well-differentiated lung cancer group was significantly higher than that of poorly differentiated lung cancer group( P0.05).Conclusion miRNA-449 a can be used as an important marker for screening lung cancer and evaluation of staging of lung cancer.
