Norepinephrine Induces Apoptosis in Human Mesangial Cells Via ROS- TNF- α Pathway

摘要 目的:探讨交感神经递质去甲肾上腺素(norepinephrine,NE)对人系膜细胞(human mesangial cells,HMC)凋亡的影响及其机制。方法:实验主要采用Tunel法检测NE对细胞凋亡的影响,采用ELESA法检测NE对肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)影响。结果:Tunel法证实NE诱导人系膜细胞凋亡通过β-肾上腺素受体(β-adrenergic receptor,β-AR),并呈浓度和时间依赖性(n=5,P<0.01,vs control组),ELESA证实NE能够诱导TNF-α分泌,并且与凋亡相关,抗氧化剂维生素C(vitamin C,VC)和N-乙酰-Ⅰ-半胱氨酸(n-acetyl-Ⅰ-cysteine,NAC)能够抑制NE诱导细胞凋亡和TNF分泌(n=5,P<0.01,vs NE组)。结论:NE通过β-AR激活和ROS-TNF-α途径诱导细胞凋亡。 Objective: Sympathetic activity is increased in the chronic renal diseases and the level of catecholamine is risen and catecholamine modulate mesangial cell function in this study, to investigate the efects of norepinephrine on the apoptosis in human mesangial cells. Methods: Investigating norepinephrine - induced the apoptosis in human mesangial ceils by Tunel; the effect of norepinephrine on Tumor necrosis factor- α( TNF -α)by ELESA. Resuits:Tunel results impilcate that NE induces apoptosis via β - ARactivation, on a concentration and time dependent manner (n =5, P 〈0.01, vs control group), ELESA datas show NE induces production of TNF - α, which mediates NE - induced apoptosis. Antioxidants including vitamin C and NAC inhibit NE - induced apoptosis ( n = 5, P 〈 0.01, vs NE group). Conclusion: NE induces cells apoptosis via β - ARactivation and ROS - TNF - α pathway.
