生长激素和胰岛素对肝组织和HepG2细胞AKT、STAT5磷酸化的调节 被引量:4

摘要 生长激素(growth hormone, GH)在机体生长和代谢方面起到重要作用, 在临床的使用日益广泛[1-4]。GH介导的Janus激酶/信号转导子和转录活化子(Janus kinase/signal transducer and activator of transcription, JAK/STAT)信号通路参与细胞的增殖、分化、迁移及凋亡等重要生物学过程[5]。GH和细胞膜上的GH受体(growth hormone receptor, GHR)相互作用后激活JAK2和GHR, 进一步活化STAT5, STAT5转移到核内与其调节基因的启动子特殊序列结合, 从而激活下游基因的转录[6]。胰岛素是一种调节代谢和生长的重要激素, 主要用于糖尿病的治疗。胰岛素与胰岛素受体(insulin receptor, IR)结合促进胰岛素受体底物(insulin receptor substrate, IRS)的磷酸化, 磷酸化的IRS和磷脂酰肌醇3激酶(phosphoinositide-3-kinase, PI3K)的调节亚基p85结合, 进一步活化PI3K。
