三七提取液对SMMC-7721细胞Bcl-2与Bax蛋白表达的影响

Effects of the Extract from Panax Notoginseng on Protein Expression of Bcl-2 and Bax in SMMC-7721 Cell Line

目的:研究三七提取液体外抑制人肝癌细胞株SMMC-7721增殖及对凋亡调控基因Bax、Bcl-2蛋白表达的影响。方法:不同浓度的三七提取液作用于体外培养的SMMC-7721细胞,采用MTT法检测细胞的抑制率;以流式细胞术观察细胞凋亡率的变化;以蛋白免疫印迹(Western blotting)法检测细胞凋亡相关因子Bcl-2及Bax和蛋白水平的表达情况。结果:三七提取液对人肝癌SMMC-7721细胞的增殖具有抑制作用,且具有明显的时间和剂量相关性。细胞凋亡率随着药物浓度的增加而逐步增高。三七提取液作用后细胞内Bcl-2蛋白表达水平降低,Bax蛋白表达水平均升高,二者的变化趋势均呈一定的时间依赖关系。结论:三七提取液能够显著抑制人肝癌细胞增殖,诱导SMMC-7721细胞凋亡,其作用机制可能与上调Bax的表达及下调Bcl-2的表达有关。

Objective： To investigate the effect of the Extract from Panax Notoginseng on proliferation,protein expression of apoptosis regulate gene such as Bax and Bcl- 2 of human liver cancer cell line SMMC- 7721 in vitro. Methods： SMMC- 7721 cells were treated with different concentrations of the Extract from Panax Notoginseng. The cells growth inhibitory rate was measured by MTT assay.Cell apoptosis rate was analyzed by flow cytometry（ FCM）. Western bloting analyses were used to detect the protein expressions of apoptosis related factors Bcl- 2 and Bax. Results： MTT methods indicated that the extract from Panax Notoginseng could inhibit the proliferation of SMMC- 7721 cells in a dose and time dependent manner. As the concentration of Panax Notoginseng extract increased,the apoptosis rate of the cells increased. After treated with the Extract from Panax Notoginseng,the protein expression of Bcl- 2 expression was down- regulated while those of Bax were up- regulated and tendency towards both changes has time- dependent relation. Conclusion： The Extract from Panax Notoginseng could inhibit the proliferation and induce cell apoptosis of SMMC- 7721 and its mechanism may be relate with its up- regulation of Bax expression as well as down- regulation of Bcl- 2 expression.
