摘要
目的:探讨迁移诱导蛋白7(migration inducing-protein 7,Mig-7)和血管生成拟态(vasculogenic mimicry,VM)在胶质瘤组织中的表达及其临床意义。方法:应用免疫组织化学法和蛋白质印迹法检测胶质瘤组织和正常对照组(外伤脑组织)中Mig-7的表达,CD34和PAS(periodic acid-Schif)双重染色法检测胶质瘤组织的VM,分析Mig-7的表达和VM与胶质瘤患者临床病理特征之间的关系,以及Mig-7的表达水平与胶质瘤患者生存期之间的关系。结果:胶质瘤组织中Mig-7的阳性表达率为56.7%(34/60),正常对照组脑组织中Mig-7的阳性表达率为0%(0/10)。Ⅰ+Ⅱ级胶质瘤组织中Mig-7的表达水平明显低于Ⅲ级和Ⅳ级胶质瘤组织(P<0.001)。胶质瘤组织中VM的阳性率为70%(42/60),正常对照组脑组织中VM阳性率为0%(0/10)。Ⅰ+Ⅱ级胶质瘤组织中的VM数明显低于Ⅲ级和Ⅳ级胶质瘤组织(P=0.003,P<0.001)。Mig-7和VM与胶质瘤患者的级别有关(P<0.001,P=0.011)。Mig-7低表达组、中表达组和高表达组患者的中位生存时间分别为85.00、36.00和10.00个月(P<0.05)。结论:胶质瘤组织中存在Mig-7的表达,其表达水平与胶质瘤的级别有关;Mig-7可能促进VM的形成,且影响患者的预后。
Objective: To investigate the expression of migration-inducing protein 7 (Mig-7) and vasculogenic mimicry (VM) in glioma and their clinical significance.
Methods: The expressions of Mig-7 in glioma tissues and normal control tissues (traumatic brain tissues) were detected by immunohistochemistry and Western blotting, respectively. The VM in glioma tissues was determined by CD34 and periodic acid-Schiff (PAS) staining method. The associations of the expression of Mig-7 and VM with the clinicalpathological characteristics of patients with glioma were analyzed. The relationship between the expression of Mig-7 and the survival of patients with glioma was also analyzed.
Results: The positive expression rates of Mig-7 in glioma tissues and the normal brain tissues were 56.7% (34/60) and 0% (0/10), respectively. The expression level of Mig-7 in grades I and Ⅱ glioma tissues was lower than that in grades Ⅲ and Ⅳ glioma tissues (P 〈 0.001). The positive expression rates of VM in glioma tissues and the normal brain tissues were 70% (42/60) and 0% (0/10), respectively. The number of VM in grades I and Ⅱ glioma tissues was smaller than that in grades Ⅲ and Ⅳ glioma tissues (P = 0.003, P 〈 0.001). The expressions of Mig-7 and VM were associated with the pathologic grade of glioma (P 〈 0.001, P = 0.011). The median survival time of patients with low, moderate and high expression levels of Mig-7 were 85.00, 36.00 and 10.00 months, respectively (P 〈 0.05).
Conclusion: The expression of Mig-7 can be observed in glioma tissues and is related to the pathologic grade of glioma. Mig-7 may promote the formation of VM and influnce the prognosis.
