紫外分光光度法测定猪血中提取免疫球蛋白IgG含量的可行性研究 被引量:2

摘要 [目的]探讨紫外分光光度法测定猪血中Ig G含量的可行性。[方法]采用饱和硫酸铵法从新鲜猪血中提取Ig G,并采用紫外分光光度法测定提取猪血中Ig G含量。[结果]将从猪血中分离的免疫球蛋白Ig G于40℃下静置40 min,p H为7.4时,获得最多Ig G的硫酸铵浓度为30%-35%。[结论]紫外分光光度法可用于测定猪血中Ig G含量,该方法数据稳定,线性关系好,准确性高,操作简便,适合推广。 [Objective] The research aimed to discuss the feasibility of determining the content of Ig G in pig blood by UV spectrophotometry.[Method]Ig G was extracted from fresh pig blood by using saturated ammonium sulphate method. And Ig G content was determined by using UV spectrophotometry. [Result] The separated Ig G extracted from pig blood was placed at p H of 7. 4 at 40 ℃ for 40 min,the highest yield of Ig G was obtained when the concentration of ammonium sulphate was 30% - 35%. [Conclusion] UV spectrophotometry could be used to determine Ig G content in pig blood. This method had the advantages of stable results,good linear relationship,high accuracy and simple operation,so it was worthy to be popularized.
