摘要
[目的]探索一种简单、科学、准确的分析多酚的方法,使富含多酚的板栗总苞废弃物得到充分的再利用。[方法]采用Folin-Ciocalteu比色法测定板栗总苞中多酚含量,并对碳酸钠、Folin-Ciocalteu试剂用量及显色温度、显色时间进行优化。[结果]该测定方法的最佳条件为:取0.50 ml板栗总苞提取液,加入3.0 ml福林-酚试剂、6.0 ml 15%的Na2CO3溶液,在40℃条件下反应70 min,测定最佳波长为760 nm。吸光度与多酚含量在0-9μg/ml范围内有良好的线性关系(y=0.135 1x+0.019 0,R2=0.999 3)。该方法的平均加标回收率为99.5%,相对标准偏差为1.2%(n=5)。[结论]该方法具有精密度高、稳定性好、操作简单、快速、结果准确等优点,可用于板栗总苞多酚含量的测定。
[Objective]To explore a simple,scientific and accurate polyphenol analysis method. [Method]The optimum condition on determining total polyphenols content in chestnut shell by Folin-Ciocalteu method was studied. [Result] The results indicated that the optimum conditions of total polyphenols determination by Folin-Ciocalteu was as follow: 3. 0 ml of Folin-phenol reagent wae added in 0. 5 ml of sample and then6. 0 ml of 15% sodium carbonate was added. After the coloration reaction was 70 min at 40 ℃,optical density was measured at 760 nm. Undder the optimum conditions,the absorbanced linearly with the optical concentration of gallic acid in the range of 0 to 9 μg / ml( y = 0. 135 1x +0. 019 0,R2= 0. 999 3). The average spiked recovery rate was 99. 5%( RSD = 1. 2%). [Conclusion]This method is highly precise and stable,and suitable for determining total polyphenol content in chestnut shell.
