EphB4、EphrinB2和HIF-1α在脑星形细胞瘤组织中的表达及意义

Expression and Significance of EphB4,EphrinB2 and HIF-1α in Brain Astrocytomas

摘要 目的探讨促红细胞生成素肝细胞(Eph)B4、Eph受体相互作用蛋白(Ephrin)B2和缺氧诱导因子-1α(HIF-1α)在脑星形细胞瘤组织中的表达水平及意义。方法采用原位杂交法检测120例脑星形细胞瘤组织标本(病例组)和20例脑白质组织标本(对照组)的EphB4、EphrinB2和HIF-1α表达水平。结果病例组EphB4、EphrinB2和HIF-1α表达水平均明显高于对照组(P<0.05),病例组病理分级Ⅲ—Ⅳ级、临床表现有出血及坏死的EphB4、EphrinB2和HIF-1α阳性表达率均显著增加(P<0.05)。EphB4、EphrinB2表达水平与HIF-1α表达水平均呈正相关(r=0.483、0.486,均P<0.05)。结论 EphB4、EphrinB2和HIF-1α在脑星形细胞瘤中的异常表达与肿瘤的恶性程度有关,EphB4与EphrinB2可能存在协同作用。 Objective To explore the expression and significance of EphB4,EphrinB2 and HIF-1αin human brain astrocytomas.Methods In situ hybridization was performed to detect the expression of EphB4,EphrinB2 and HIF-1αin 120 brain astrocytoma specimens(case group)and 20 cerebral white matter specimens(control group).Results Compared with control group,the expression of EphB4,EphrinB2 and HIF-1αsignificantly increased in case group(P〈0.05).Pathologic stageⅢ-Ⅳ grade,clinical manifestations of hemorrhage and necrosis in cose group,the positive expression rate of EphB4,EphrinB2 and HIF-1αsignificantly increased(P〈0.05).The expression of EphB4 and EphrinB2 was positively correlated with the expression of HIF-1α(r=0.483 and 0.486,respectively;P〈0.05).Conclusion The overexpression of EphB4,EphrinB2 and HIF-1αin brain astrocytomas is associated with the malignant degree of tumors.EphB4 and EphrinB2may be synergies.
