川芎-天麻不同比例配伍药效成分含量变化的研究 被引量:4

摘要 目的建立HPLC法测定川芎-天麻药对70%(体积分数,下同)乙醇提取液中药理活性成分阿魏酸、天麻素和天麻苷元的质量浓度,研究药对中不同比例配伍对目标组分的影响。方法采用Waters e2695高效液相色谱仪、GL Science Inertsil ODS-3色谱柱(250 mm×4.6 mm,5μm);柱温25℃,流速1 m L·min-1。阿魏酸检测的流动相为甲醇-0.05%(体积分数)H3PO4水溶液(体积比40∶60),波长为320nm;天麻素、天麻苷元检测的流动相为乙腈-0.05%(体积分数)H3PO4水溶液(体积比3∶97),波长为220nm。结果用本法测定得到不同比例配伍的川芎-天麻药对70%乙醇提取液中各指标成分质量浓度随药材比例改变而改变。结论本试验建立的HPLC法可有效测定不同比例配伍的川芎-天麻药对的70%乙醇提取液中阿魏酸、天麻素和天麻苷元的质量浓度,可为川芎-天麻药对体内外的成分研究和临床运用提供参考。 Objective An HPLC method was established for quantitative analysis of ferulic acid,gastrodin and p-hydroxybenzyl alcohol in decoction with Chuanxiong Rhizoma( CR)-Gastrodiae Rhizoma( GR),and to investigate the effect of CR and GR in different proportions on the yield of the components above.Methods Waters e2695 was used and HPLC was carried out with a GL Science Inertsil ODS-3( 250 mm×4. 6 mm,5 μm) colomn. The colomn temperature was 25 ℃ and the flow rate was 1 m L·min^-1. Methanol-0.05% phosphoric acid( 40 ∶ 60) was used as mobile phase for ferulic acid and acetonitrile-0. 05%phosphoric acid( 3 ∶97) for gastrodin and p-hydroxybenzyl alcohol. Detection wavelengths were 320 nm for ferulic acid and 220 nm for gastrodin and p-hydroxybenzyl alcohol. Results The concentrations of three substances in the decoction varied with different proportions of CR and GR. Conclusion The HPLC method was suitable for the determinations of ferulic acid,gastrodin and p-hydroxybenzyl alcohol in decoction with CR and GR,which provided reference for its clinical use and further research in vivo and in vitro.
