摘要
目的:探讨清心开窍方有效成分对AD大鼠海马区IL-1β、GFAP及Aβ表达的影响。方法:动物实验采用对照观察法。雄性SD大鼠112只,称体质量后按随机数字表分为7组:正常组、假手术组,模型组、安理申组,皂苷组、挥发油组、多糖组,每组16只。采用双侧海马注射Aβ1-40制造AD大鼠模型,造模后第2天开始灌胃,连续灌胃2周(每天上午10点开始),清心开窍方皂苷组、清心开窍方挥发油组、清心开窍方多糖组剂量分别是9、3.33、8.33 m L/(kg·d),安理申组给药剂量为1.67 mg/(kg·d),予对照组和模型组等体积生理盐水。2周后,采用免疫组化、实时定量荧光PCR及WB法检测AD大鼠海马区IL-1β、GFAP及Aβ表达。结果:清心开窍方各有效成分组大鼠海马区IL-1β、GFAP及Aβ表达水平降低,与对照组比较差异非常显著(P<0.01,P<0.05)。结论:清心开窍方有效成分可能是通过降低海马区IL-1β、GFAP及Aβ的表达发挥抗AD的作用。
Objective: To explore the effect of the active ingredient of Qingxin Kaiqiao Prescription on IL- 1β,GFAP and Aβ expressions in hippocampus of AD model rats. Methods: Animal experiment used comparison method. 112 male SD rats were rando m Ly divided into seven groups: normal,control,model,Aricept,saponin,benzene and polysaccharide groups. Each group had sixteen rats. Alzheimer's disease rat model was produced by bilateral hippocampus injection of Aβ1- 40. The next day after modeling,rats were treated with saponin [9 m L /( kg·d) ] or polysaccharide [3. 33 m L /( kg·d) ]or polysaccharide [8. 33 m L/( kg·d) ]or Aricept [1. 67 mg/( kg·d) ]once a day for consecutive 2 weeks( at 10 a. m. every day),separately. Control group and model group were given volume-matched saline. After 2 weeks,immunohistochemistry,real time-PCR and western bolting methods were used to detect the expressions of IL- 1β,GFAP and Aβ in hippocampus of AD rats. Results: Levels of IL- 1β,GFAP and Aβ in hippocampus of the active ingredients( spaonin,benzene and polysaccharide) of Qingxin Kaiqiao Prescription were decreased compared to those of the control group( P〈 0. 01,P 0. 05). Conclusion: The active ingredients of Qingxin Kaiqiao Prescription may treat Alzheimer's disease by decreasing the expressions of IL- 1β,GFAP and Aβ in hippocampus.
