摘要
目的:观察络治法对链佐脲菌素(STZ)诱导的糖尿病大鼠视网膜血管消化铺片中血管细胞粘附分子-1(VCAM-1)表达的影响,探讨络治法对糖尿病视网膜病变微血管损伤的保护作用及机制。方法:60只雌性Wistar大鼠随机分成空白组、模型组、羟苯磺酸钙对照组及络治组4组。除空白组外各组均制作成糖尿病大鼠模型。给药24周后每组大鼠取双侧眼球做视网膜血管消化铺片,右眼行PAS染色做形态学观察,左眼免疫荧光染色法测定VCAM-1在血管壁中的表达。结果:PAS染色结果显示,络治组糖尿病大鼠视网膜毛细血管在管径粗细、局部扩张程度、扭曲聚集程度等方面较对照组及模型组有明显的改善,且络治组管壁周细胞的损伤明显减少,组间差异有统计学意义(P<0.05)。免疫荧光染色显示,络治组视网膜血管壁中VCAM-1呈较少的表达,组间差异有统计学意义(P<0.05)。结论:络治法能够抑制VCAM-1在糖尿病视网膜毛细血管壁的表达,有效减轻高血糖引起的视网膜微血管损伤,改善糖尿病视网膜微血管的结构与机能。
Objective: To observe vascular cell adhesion molecule in- 1( VCAM- 1) expression in the retinal digest preparations of streptozotocin( STZ) induced diabetic rats treated with collaterals therapy,and to investigate the protective effects and mechanism of collaterals therapy on diabetic retinopathy microvascular injury. Methods: 60 female wistar rats were randomly divided into 4 groups including normal group,model group,calcium dobesilate group and collaterals therapy group. All except the normal group rats were made into models of diabetic rats. After 24 weeks each rat's bilateral eye were made into retinal vascular digest preparations,PAS staining for morphological observation of the right eye,the left eye for immunofluorescence staining method to determine the VCAM- 1 expression in the vessel wall. Results: PAS staining showed,collaterals therapy group had obvious improvement on morphological changes such as vessel diameter,the local expansion、vascular twist and so on. However pericyte damage of collaterals therapy group was significantly suppressed,comparison between groups were statistically significant( P〈 0. 05). Immunofluorescence staining showed that vcam- 1 in collaterals therapy group has less expression than other group,comparison between groups were statistically significant( P〈 0. 05). Conclusion: Collaterals therapy can inhibit the expression of VCAM- 1 in diabetic retinal capillary wall,effectively alleviate the hyperglycemia induced retinal microvascular damage,improve diabetic retinal microvascular structure and function.
