摘要
目的:探讨HIF-1α、MMP-2、MMP-9等易感基因在非小细胞肺癌(NSCLC)癌组织中的表达水平。方法:98只通过化学诱导的方法诱导为NSCLC模型的SD大鼠,作为观察组;同时选择60只肺部良性病变的SD成年雄性大鼠作为对照组。测定上述易感基因在良性组织以及肿瘤组织中的阳性表达率,以及随着病情的加重上述基因阳性表达率的变化情况。HIF-1α表达采用HIF-1α试剂盒对单克隆抗体HIF-1α进行检测;MMP表达采用免疫组化的方法在高倍显微镜下进行检测。结果:HIF-1α、MMP-9、MMP-2在对照组组织中阳性表达率分别为5.0%、8.3%、10.0%,在观察组中阳性表达率分别为79.6%、78.6%、74.5%(P均<0.05)。随着病情的加重HIF-1α、MMP-9、MMP-2在肿瘤组织的阳性表达率有逐渐升高的趋势。结论:与良性组织相比,HIF-1α、MMP-9、MMP-2在肿瘤组织中显著高表达,因此有必要对上述基因进行进一步研究。
Objective:To investigate the gene expression of HIF-1α,MMP-2,MMP-9 and other predisposing in non-small cell lung cancer( NSCLC).Methods:All 98 SD rat model,were used as the observation group,60 benign lung lesions adult male SD rats were used as controls.HIF-1α expression was detected by HIF-1α kit monoclonal antibodies.MMP expression was detected using immunohistochemical methods.Results:HIF-1α,MMP-9,MMP-2 in the control group,the positive expression rates were 5.0%,8.3%,10.0%,HIF-1α,MMP-9,MMP-2positive expression rates in the observation group were 79.6 %,78.6%,74.5%( P〈0.05).As the disease devel oped HIF-1α,MMP-9,MMP-2 expression rate in tumor tissue has gradually increased.Conclusion:Compared with healthy tissue,HIF-1α,MMP-9,MMP-2 have a significantly higher expression in tumor tissue.
