HAART对HIV-1感染者CD_4^+CD25^+FoxP3^+调节性T细胞构成比活化和稳态的影响 被引量:3

The effects of HAART on the frequency,activation and homeostasis of CD_4^+CD25^+FoxP3^+ regulatory T cells

摘要 目的观察高效抗反转录病毒治疗(HAART),对艾滋病病毒Ⅰ型(HIV-1)感染者外周血CD+4CD25+FoxP3+调节性T细胞(Treg)的构成比、活化和更替水平的影响。方法对入组的17例慢性HIV-1感染病人进行6个月HAART治疗,收集治疗前及治疗6个月后病人外周血,检测外周血调节性T细胞的构成比及CD38、人白细胞抗原(HLA)-DR等活化标志,同时检测调节性T细胞增殖标志Ki-67和凋亡标志Caspase-3。结果 HAART完全应答者外周血中的Treg构成比、CD38和HLA-DR的表达、Ki-67和Caspase-3的表达均较无应答者有显著降低。结论 HAART能够抑制调节性T细胞活化和更替,这有助于进一步了解HAART的免疫学机制。 Objective To investigate the effects of highly active anti-retroviral therapy(HAART)treatment on the frequency,activation and homeostasis of peripheral blood CD^+4CD25^+FoxP3^+ regulatory T cells from chronic human immunodeficiency virus-1(HIV-1)infected patients.Methods Seventeen chronic HIV-1infected patients who received HAART treatment were enrolled and followed up for six months.Peripheral blood was collected before and after six-month HAART treatment,and the frequency of CD+4CD25+Fox3+regulatory T cells was investigated.In addition,the expression of activation markers-CD38,HLA-DR and homeostatic markers-Ki-67 and caspase-3on Treg were also investigated.Results After six-month HAART treatment,the frequency of Treg and the expression of CD38,HLA-DR,Ki-67 and caspase-3on Treg were decreased in complete responders but not in non-responders.Conclusion HAART treatment could decrease the activation and turnover,which supply further understanding of the immunological mechanism of HAART treatment.
