摘要
目的:探讨复方芩柏颗粒剂对溃疡性结肠炎(UC)实验大鼠的治疗作用及可能的作用机制。方法:采用TNBS法制备UC大鼠模型,将造模成功的大鼠随机分为模型组、奥沙拉嗪组和复方芩柏颗粒剂组,每组20只;另选取20只健康大鼠为正常组,各组分别灌肠给药,连续3周。镜下观察结肠组织病理切片并进行病理学分级,免疫组化检测NF-κB p65的表达。结果:与正常组比较,模型组NF-κB p65的表达及病理分级评分明显升高,差异具有统计学意义(P<0.01);与模型组比较,西药组、中药组NF-κB p65的表达及病理分级评分均明显降低,差异具有统计学意义(P<0.01);西药组和中药组组间比较,差异无统计学意义(P>0.05)。结论:复方芩柏颗粒剂保留灌肠对UC实验大鼠有良好的治疗作用,其可能是通过抑制NF-κB p65表达的上调来发挥作用。
Objective: To investigate the therapeutic effect and action mechanism of compound Qinbo granules in the treatment of experimental rats with ulcerative colitis( UC). Methods: A rat model of UC was established by the 2,4,6-trinitrobenzene sulfonic acid method. After the model was successfully established,60 UC model rats were equally and randomly divided into model group,olsalazine group( Western medicine group),and compound Qinbo granule group( traditional Chinese medicine group)( n = 20 for each). And another 20 healthy rats were selected as normal group. The olsalazine group and compound Qinbo granule group were given olsalazine and compound Qinbo granules,respectively,by enema; the model group was given saline by enema; the normal group was fed conventionally. After 3 consecutive weeks of treatment,the pathological sections of colon tissue were observed under a microscope for pathological classification. The expression of nuclear factor- kappa B p65( NF- κB p65) was determined by immunohistochemistry. Results: The model group had significantly increased expression of NF- κB p65 and pathological grading score than the normal group( P〈0. 01). Compared with the model group,the Western medicine group and the traditional Chinese medicine group had significantly reduced expression of NF- κB p65 and the pathological grading score( P〈0. 01). There were no significant differences in these parameters between the Western medicine group and the traditional Chinese medicine group( P〉0. 05). Conclusion: The retention enema with compound Qinbo granules has a good therapeutic effect for the treatment of experimental rats with UC,which may be attributed to inhibiting the upregulation of NF- κB p65.
