摘要
基于毛细管电泳技术,结合间接紫外检测法,探索了4种脂肪族生物活性胺(尸胺、腐胺、精胺及亚精胺)的分离方法。文章讨论了间接紫外检测背景电解质及其浓度的选择,研究了缓冲体系p H的影响,并考查了不同种类的添加剂及其浓度对分离的作用,最终总结出最优分离条件为10 mmol·L-1咪唑-醋酸(p H=4.50)的缓冲体系,含0.5 mmol·L-1 EDTA,分离电压+20 k V,检测波长200 nm。该方法无需衍生,最佳条件下可在5 min内使4种物质基线分离,其检出限在0.9-3.2μmol·L-1,出峰时间和峰面积的重现性分别在0.28%~0.35%,2.53%~4.52%之间,只一种是分离检测食物和人体中重要生物胺的快速、方便和准确的毛细管区带电泳法。
The paper based on capillary zone electrophoresis(CZE) technology, combined with indirect-UV determination method, explored separation and determination method of 4 biogenic amines(BAs)(spermine, spermidine, cadaverine and putrescine). The paper discussed the choice of the background electrolyte and its concentration under indirect-UV determination method, analysted the influence of p H of buffer system, researched the effect of the different kinds of additives and their concentration to separation and determination, finally summed up the optimum separation condition of the buffer system of imidazole-acetic acid(p H=4.50)at 10 mmol·L-1, containing 0.5 mmol·L-1 EDTA, separation voltage at +20 k V, detection wavelength at 200 nm. The method doesn't need the derivatization. Under the optimal condition, the baseline separation of four BAs can be achieved within 5 min, its LODs was in the range of 0.9~3.2 μmol·L-1, the RSD of the peak time and peak area were in the range of 0.28 %~0.35 % and 2.53 %~4.52 %, respectively. So this is a simple, rapid and accurate capillary zone electrophoresis(CZE)method for separation and determination of biogenic amines(BAs), which widely spread and play key biological roles in the human body and foods.
