β-谷甾醇对T47D细胞雌激素受体表达及其下游基因PS2的影响 被引量:9

摘要 目的:观察β-谷甾醇对T47D细胞雌激素受体(estrogen receptor,ER)及下游基因PS2的影响,探讨其植物雌激素样作用机制。方法:以ER拮抗剂ICI182 780为工具药,应用实时荧光定量PCR技术检测ERα、ERβ及PS2mRNA表达变化,western-blot技术检测ERα、ERβ蛋白表达变化。结果:β-谷甾醇可诱导ERα、ERβ及PS2 mRNA表达,且上调ERβ更显著,对PS2 mRNA的诱导作用可以被ICI182 780抑制,能够上调ERα和ERβ蛋白表达,且可以被ICI182 780抑制。结论:β-谷甾醇可以通过ER途径调节下游靶基因的表达而发挥植物雌激素样作用。 Objective: To observe the effect of β-sitosterol on ER and downstream gene PS2 expression in breast cancer cell line T47 D cells,and explore its phytoestrogenic mechanism. Methods: mRNA expressions of ERα,ERβ and its downstream gene PS2 in T47 D were assessed by RT-PCR,protein expressions of ERα、ERβin T47 D were assessed by western-blot. ICI182 780 was employed to observe the influence on the gene expressions of PS2 and protein expressions of ERα,ERβ. Results: β-sitosterol up-regulated ERα,ERβmRNA expression. PS2 gene are induced in response to β-sitosterol treatment. The effect on PS2 gene expression was inhibited by pure estrogen receptor antagonist ICI182780. β-sitosterol increased the level of T47 D cells ERα,ERβ proteins expression,respectively. The effect was completely antagonized by ICI182 780. Conclusion: β-sitosterol up-regulated downstream gene PS2 expression through ER. And β-sitosterol possessed phytoestrogenic effect by activating ER.
