摘要
[目的]建立高效液相色谱测定烟筋中的烟碱含量。[方法]用4%Na OH溶液提取,正己烷和20%H2SO4溶液萃取,Ba(OH)2沉淀的方法提取烟筋中烟碱;选用Hypersil BDS-C18色谱柱,流动相为甲醇-水(0.02 mol磷酸二氢钾加0.52%三乙胺,用磷酸调整p H值为6.5)(体积比60颐40),检测波长260 nm,流速1.0 m L/min,柱温40℃,进样量10滋L。[结果]烟碱质量浓度在100∽1 000 mg/L时,线性关系良好;添加水平为8.00∽32.00 mg时,回收率为98.37%--99.03%,RSD为0.65%。[结论]该方法能准确快速检测烟筋中烟碱的含量。
[Aims] A HPLC method for determination of nicotine in tobacco vein was established. [Methods] The nicotine in tobacco vein was extracted with Na OH solution 4%, normal hexane and H2SO4 solution 20%, and the H2SO4 in extracts was precipitated by Ba(OH)2crystal. The nicotine in tobacco vein was determinated on Hypersil BDS-C18 chromatographic column at 40 ℃ by using methanol/water(with 0.02 mol of potassium dihydrogen phosphate and triethylamine 0.52%, and phosphoric acid was used for adjusting p H to 6.5)(60:40, by vol) as mobile phase with the flow of 1.0 m L/min, in addition, UV detection was conducted at 260 nm for 10 滋L of test sample. [Results] The results showed that the calibration curve was in good linear with nicotine content in the range of 100-1 000 mg/L. When the addition levels were 8.00-32.00 mg, the recovery rates were 98.37-99.03% with the RSD of 0.65%. [Conclusions] This method is simple with high precision and accuracy, which can be used for detection of nicotine in tobacco vein.
