Wnt3a及Lgr5在膀胱癌中的表达及意义

摘要 目的探讨Wnt3a、富含亮氨酸重复序列G-蛋白耦联受体(Lgr5)在人膀胱癌组织中的表达和意义。方法应用免疫组织化学法检测11例正常膀胱组织和39例膀胱癌组织Wnt3a、Lgr5蛋白表达情况,RT-PCR方法检测9例正常膀胱组织和17例膀胱癌组织中Wnt3a、Lgr5 mRNA的表达情况。结果免疫组化结果显示:Wnt3a蛋白在膀胱癌与正常膀胱组织表达阳性率分别为79.49%和36.36%,差异有统计学意义(P<0.05);Lgr5蛋白在膀胱癌与正常膀胱组织表达阳性率分别为74.36%和27.27%,差异有统计学意义(P<0.05);两者在膀胱癌中的表达呈正相关(r=0.677,P<0.01)。RT-PCR结果显示:Wnt3a在膀胱癌和正常膀胱组织的光密度比值分别为(0.197±0.029)和(0.088±0.019),差异有统计学意义(P<0.05);Lgr5在膀胱癌和正常膀胱组织的光密度比值分别为(0.408±0.061)和(0.161±0.053),差异有统计学意义(P<0.05)。结论在膀胱癌组织中Wnt3a、Lgr5蛋白及mRNA的表达均高于正常膀胱组织,提示Wnt3a、Lgr5可能与膀胱癌的发生发展密切相关。 Objective To investigate the expression and significance of Wnt3 a and Lgr5 in bladder cancer. Methods The expression levels of Wnt3 a protein and Lgr5 protein wee detected by immunohistochemistry in 11 cases of normal tissues and 39 cases of bladder cancer tissues. The expression levels of Wnt3 a mRNA and Lgr5 mRNA were detected by RT-PCR in 9cases of normal bladder tissues and 17 cases of bladder cancer tissues. Results The results by immunohistochemistry showed the positive expression rates of Wnt3 a protein were 84. 6%,33. 3% in bladder cancer tissues and normal bladder tissues,respectively,there were significant differences between the two kinds of tissues( P〈0. 05). The positive expression rates of Lgr5 protein were 84. 6%,33. 3% in bladder cancer tissues and normal bladder tissues,respectively,there were significant differences between the two kinds of tissues( P〈0. 05). The expression of Wnt3 a was positively correlated to that of Lgr5 in bladder cancer tissues( r = 0. 677,P〈0. 01). The results by RT-PCR showed that there was a significant difference in the ratio of optical density of Wnt3 a between bladder cancer tissues and normal bladder tissues( P〈0. 05),there was a significant difference in the ratio of optical density of Lgr5 between bladder cancer tissues and normal bladder tissues( P〈0. 05). Conclusion The expression levels of the protein and mRNA of Wnt3 a and Lgr5 in bladder cancer tissues are much higher than those in normal bladder tissues,which suggest that Wnt3 a and Lgr5 may be closely related with the pathogenesis and development of bladder cancer.
