摘要
目的检测内皮素-1(EDN-1)基因在乳腺癌患者外周血中的表达情况,并探讨它在乳腺癌的发生、发展中的作用。方法选取2013年1月1日至2014年5月20日在花都区人民医院接受乳腺手术且经病理学证实为乳腺癌的患者54例纳入试验组,另选取同期体检健康女性60例纳入对照组。采用实时定量聚合酶链反应(PCR)检测所有研究对象外周血EDN-1的表达情况,并分析其与各临床参数的关系。结果乳腺癌患者外周血EDN-1表达水平高于体检健康女性,组间比较差异有统计学意义(P<0.01),其表达水平为健康女性人群的5.877倍;EDN-1的表达水平与患者的年龄和肿瘤的组织类型均无关(P>0.05),与患者的肿瘤大小、区域淋巴结转移情况、是否有远处转移密切相关(P<0.05)。结论 EDN-1基因与乳腺癌的发生、发展关系密切,实时定量PCR可以敏感、特异地检测乳腺癌患者外周血中EDN-1的表达,可以为乳腺癌的筛查与早期诊断提供帮助,宜在临床工作中开展。
Objective To detect the expression of endothelin-1(EDN-1)gene in peripheral blood of the patients with breast carcinoma,and to investigate its role in the occurrence and development of breast carcinoma.Methods A total of 54 patients with pathologically confirmed breast carcinoma receiving operation in our hospital from1 st Jan.2013 to 20th May 2014 were selected as the experimental group.Other 60 healthy women of physical examination were selected as the control group.The real-time quantitative PCR was adopted to detect the expression of EDN-1gene in peripheral blood of all objects.The relationships between the EDN-1gene expression and clinicopathological parameters were analyzed.Results The expression of peripheral blood EDN-1gene in the patients with breast carcinoma was significantly higher than that in healthy women,the difference was statistically significant(P〈0.01),and the expression level of EDN-1gene was 5.877 times higher than that of healthy women.The expression level of EDN-1had no correlation with the age of patients and the pathological types of breast carcinoma(P〉0.05),while had close correlation with the tumor size,regional lymph node metastasis and distant metastasis(P〈0.05).Conclusion EDN-1gene might closely correlated with the occurrence and development of breast carcinoma.The real-time quantitative PCR could sensitively and specifically detect the peripheral blood expression of EDN-1gene in breast carcinoma,which might be helpful for the screening and early diagnosis of breast carcinoma and could be carried out in clinical practice.
