摘要
目的探讨细胞色素P450 3A4(CYP3A4)基因多态性与中国汉族女性乳腺癌易感性的相关性。方法用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)的方法检测148例中国汉族女性乳腺癌患者及160例健康受试者的CYP3A4*1G多态性,分析2组CYP3A4*1G基因型及等位基因分布频率。结果中国汉族女性乳腺癌患者和健康受试者CYP3A4*1G基因型分布均符合Hardy-Weinberg平衡。乳腺癌患者和健康受试者CYP3A4*1G等位基因频率分别为15.9%和22.2%(P<0.05),CYP3A4*1G基因型的分布频率在2组间差异有统计学意义(P<0.01)。结论 CYP3A4*1G多态性与中国汉族女性乳腺癌的易感性存在负相关性。
Objective To detect the frequency distribution patterns of cytochrome P450 3A4 (CYP3A4) polymorphism in Chinese Han female healthy volunteers and patients with breast cancer. Methods A case - control study was carried out to compare the distribution of genotypes and genetic frequencies of CYP3A4 * 1G between breast cancer patients ( n = 148 ) and healthy volunteers ( n = 160 ) by polymerase chain reac- tion- restriction fragment length polymorphism (PCR- RFLP). Results The distribution of genotypes and genetic frequencies of CYP3A4 * IG were both in accordance with Hardy -Weinberg balances. The frequencies of CYP3A4 * 1G allele in the cases and controls were 15. 9% and 22. 2%, respectively (P 〈 0. 05). There was a significant difference of CYP3A4 * 1G genotypes between the two groups ( P 〈 0. 01 ). Conclusion The polymorphism of CYP3A4 * 1G was inversely correlated with the suscepti- bility of breast cancer.
