荷丹片对APOE^(-/-)小鼠血脂、炎症因子及形态学变化的影响 被引量:9

摘要 目的研究荷丹片(荷叶,丹参,山楂,番泻叶,补骨脂)对载脂蛋白E基因敲除(APOE-/-)小鼠的血脂、脂联素、肿瘤坏死因子(TNF-α)、高敏C反应蛋白(hs-CRP)和形态学变化的影响。方法将50只APOE-/-小鼠随机分为对照组、模型组、荷丹片低、高剂量组、辛伐他汀组。对照组普通饲料喂养,其余各组高胆固醇饲料喂养。12周后,取血清测血脂,ELISA法测脂联素、TNF-α、hs-CRP水平;取同部位主动脉HE染色光镜下观察形态学变化。结果与对照组相比,模型组血脂、TNF-α、hs-CRP水平明显升高(P<0.01),脂联素水平明显降低(P<0.01),主动脉形态学病变加重。与模型组相比,荷丹片低、高剂量组、辛伐他汀组的血脂、TNF-α、hs-CRP水平明显降低(P<0.01),脂联素水平明显升高(P<0.05),主动脉形态学病变有所减轻。结论荷丹片可降低APOE-/-小鼠血脂和TNF-α、hs-CRP水平,升高脂联素水平,从而抑制动脉粥样硬化的发生、发展。 AIM To investigate the effects of Hedan Tablets( Nelumbinis Folium,Salviae miltiorrhizae Radix et Rhizoma,Crataegi Fructus,Sennae Folium,Psoraleae Fructus) on serum lipid,adiponectin,TNF-α,hs-CRP and morphological changes of APOE^-/- mice.METHODS Fifty APOE^-/- mice were randomly divided into normal control group,the model group,Hedan Tablets low and high dose groups and Simvastatin group.Mice in the control group were given ordinary chow and mice in other groups were given high cholesterol diet.After twelve weeks,mice were sacrificed to measure serum lipid.Adiponectin,TNF-α,hs-CRP were measured by ELISA.Morphological changes in aorta were observed under the light microscope.RESULTS Compared with the control group,the serum lipid,TNF-α,hs-CRP significantly increased( P〈0.01),adiponectin significantly decreased( P〈0.01) and aortic morphology lesions were serious in the model group.Compared with the model group,the serum lipid,TNF-α,hs-CRP significantly decreased( P〈0.01),adiponectin significantly increased( P〈0.05)and aortic morphology lesions were reduced in Hedan Tablets groups and Simvastatin group.CONCLUSION Hedan Tablets can reduce the levels of serum lipid,TNF-α,hs-CRP while increase adiponectin level of APOE- /-mice,and then inhibit the formation of atherosclerosis.
