摘要
采用Alcalase对黄斑海蜇蛋白进行酶解,以水解度为指标,通过正交试验法对酶解工艺进行优化,确定了最佳酶解工艺参数为酶与底物为5%,酶解时间为4.0 h,酶解p H为8.0,酶解温度为60℃。在此条件下,水解度达到33.87%。体外生物活性测定显示,黄斑海蜇蛋白酶解产物具有较强的DPPH自由基,羟基自由基和超氧阴离子清除能力,和显著的血管紧张素转化酶(ACE)抑制活性,为黄斑海蜇的开发利用提供理论依据。
Enzymatic hydrolysis of Rhopilema hispidum vanhoffen and its biological activities in vitro were investigated. Alcalase was selected to hydrolyse R. hispidum. Using the degree of hydrolysis (DH) as the index, orthogonal design experiment was employed to optimize the enzymatic hydrolysis process. The optimum parameters were as followings: enzyme/subslrate ratio 5%, pH 8.0, hydrolysis time 4.0 h and hydrolysis temperature 60 ℃. Under this condition, DH reached to 33.87%. In vitro biological activities investigation demonslrated that R. hispidum protein hydrolysate possessed potential DPPH, hydroxyl radicals and superoxide anions scavenging activities, and it also exhibited notable angiotensin converting enzyme inhibitory activity. This study provided theoretical basis for development ofR. hispidum.
