摘要
采用固相萃取(SPE)-高效液相色谱法(HPLC)检测羊肉风干肠中色胺、尸胺、组胺、酪胺、亚精胺和精胺6种生物胺(BAs)。对比了液液萃取和采用不同萃取柱(ODS-C18萃取柱、阳离子SCX萃取柱、阴离子萃取SAX柱)进行固相萃取对样品中生物胺的净化效果。通过研究发现,固相萃取法对生物胺具有较好的净化、富集作用,其中强阳离子SCX萃取柱效果最佳,生物胺线性回归范围在0.5~25 mg/L,方法的检出限在0.01~0.1 mg/L之间,定量限在0.1~1.0 mg/L之间。羊肉风干肠中加标回收率为74.25%~98.37%,相对标准偏差(RSD,n=6)为9.36%~17.27%,方法的准确性和回收率较高,可以有效检测羊肉风干肠中生物胺含量。
A method for the determination of tryptamine,cadaverine,histamine,tyramine,spermidine,spermine in mutton dried sausage by solid phase extraction(SPE) and high performance liquid chromatography(HPLC) was established.The purification effects of liquid-liquid extraction and solid phase extraction with different cartridges(ODS-C(18),SCX,SAX) had been compared,the solid phase extraction showed a higher recovery,the strong cationic exchange(SCX) cartridge had the highest recovery.The method was linear for the amines studied at concentration ranging from 0.5 to 25 mg/L,the limits of detection of the method were between 0.01 and 0.1 mg/L,the limits of quantification were between 0.1 and 1.0 mg/L.The average recoveries of samples with standard solution ranged from 74.25% to 98.37% for all amines,the relative standard deviations(RSDs,n=6) were in the ranges of 9.36%~17.27%.This method has a higher accuracy and recovery.It can be used for the determination of biogenic amines in mutton dried sausage.
