摘要
通过凯氏定氮法测定盐肤木种子总蛋白质含量,然后采用3种不同的方法提取其中的蛋白质。结果表明:盐肤木种子总蛋白质的含量约为9.501%。在本实验条件下,改良丙酮沉降法提取的蛋白质含量较TCA-丙酮提取法和碱式提取法高。正交试验确定改良丙酮沉降法较优工艺条件为:"料液比为1∶12g·mL-1,提取时间为3h,提取液与沉降蛋白所用丙酮量体积比1∶2,沉降时间为1.5h。在较优提取工艺条件下,盐肤木蛋白质的提取率达到58.44%。
The content of protein from Rhus Chnensis seed was assayed by Kjeldahl determination and three pro- tein extraction methods were employed. The results showed that the content of Rhus Chnensis seed protein was about 9. 501%. Protein extracted by the improved acetone sedimentation method was higher than by the TCA --acetone extraction and basic extraction method. The optimum process conditions of improved acetone sedi- mentation method by orthogonal test were: the ratio of solid to liquid 1 : 12 g · mL^-1 ,extraction time 3 h,the ratio of protein extraction solution to acetone 1 : 2,sedimentation time 1.5 h. Practicing this optimal condition, the extraction rate of Rhus Chinensis protein reached 58.44%. The experiment data was referred for further study of the Rhus Chinensis seed protein.
