摘要 Actinoplanes sp.N902-109是继吸水链霉菌ATCC292953后分离的第二株能够产生雷帕霉素的放线菌。采用Roche 454GS FLX测序技术和Sanger法PCR测序技术,我们首次完成了Actinoplanes sp.N902-109全基因组序列测定。Actinoplanes sp.N902-109基因组为环状,长度为9228054bp,GC含量71.3%,编码8212个蛋白,其中5460个编码蛋白被注释有明确的生物学功能。应用Anti SMASH和NRPS predictor软件预测基因组中存在22个次级代谢生物合成基因簇,包含首次鉴定的完整的游动放线菌来源雷帕霉素生物合成基因簇。利用Mummer和Mauve软件对N902-109和已公布的游动放线菌属Actinoplanes SE50/11 0和Actinoplanes missouriensis 431两株菌株全基因组进行比较分析。通过Actinoplanes sp.N902-109基因注释和比较基因组学分析,在全基因组水平上了解Actinoplanes sp.N902-109的遗传物质基础,为开展Actinoplanes sp.N902-109的代谢调控研究和遗传重组改造奠定基础。 Actinoplanes sp. N902-109 is a soil bacterium known for its high titer ofrapamycin, a pharmaceutically important immunosuppressant. Here, we present the genome sequence of it which was generated using a shotgun approach employing 454 sequencing technologies. Gaps between contigs were closed by PCR primer walking. The circular genome ofActinoplanes sp. N902-109 was 9228054bp in length with GC content of 71.3%. A total of 8,212 open reading frames(ORF) were predicted, in which 5,460 ORFs were annotated with clear biological functions. By using NRPS Predictor and AntiSMASH gene prediction software, 22 putative biosynthetic gene clusters were found in the genome, including the complete rapamycin biosynthetic gene cluster which was identified for the first time in this bacterium. This genome was compared with the published genomes of two Actionoplanes species by Mummer and Mauve softwares. The genome sequence ofActinoplanes sp. N902-109 has been deposited in GenBank under accession no: CP005929.1. The availability of the genome sequence of Actinoplanes sp. N902-109 allows for understanding the biosynthesis ofrapamycin and will facilitate rational genome modification to improve industrial use.
