摘要
目的研究大孔吸附树脂分离纯化非达霉素的工艺条件。方法以非达霉素的吸附量为考察指标,从中筛选树脂;并对吸附及解吸条件进行优化,得到适宜的纯化操作条件。结果 HZ816树脂为最佳吸附剂,优化的条件为吸附流速为2BV/h,解吸剂为80%的乙醇-水溶液。在此实验条件下所得产品纯度≥90%,收率≥60%。结论 HZ816树脂对非达霉素有较好的吸附分离性能,适合于非达霉素的提纯。
Objective To study technological parameters of fidaxomicin separation and purification with macroporous adsorption resin. Methods The adsorption capacity offidaxomicin was used as index toselect the resin; and the optimal purification process was determined by improving the adsorption and desorption conditions. Results Resin HZ816 was selected as the best adsorbent, the optimum condition was investigated as follows: adsorption flowrate of 2BV/h, 80% ethanol-H20 as eluent. Under the optimum condition the purity of the final product was over 90% and the yield of fidaxomicin was above 60%. Conclusion Resin HZ816 showed a perfect separation performance on fidaxomicin and was suitable to purify it.
