摘要
[目的]研究猕猴桃实生苗培育与砧木的工厂化生产技术。[方法]猕猴桃种子通过GA3不同处理后进行组织培养。[结果]猕猴桃种子通过2.5g/LGA3浸种12 h,用10%次氯酸钠消毒10min,播种于铺有一层湿润滤纸的培养皿中进行暗培养,以获得较高的发芽率。将发芽的种子及时转入培养基(MS+2 mg/L 6-BA+0.05 mg/L NAA+30g/L蔗糖+7g/L琼脂粉,pH 5.8)中,在组培室内进行培养,可迅速获得猕猴桃实生苗。以带芽茎段为外植体对猕猴桃实生苗进行单株扩繁,可得到基因型相同的株系。[结论]‘徐香’和‘布鲁诺’实生苗可以作为猕猴桃的砧木使用,以实现猕猴桃砧木的工厂化生产。
[ Objective ] To study the technique of cultivating seedling and rootstock' s factory production of kiwifruit. [ Method ] Kiwifruit seeds were used as materials in tissue culture after different treatments with GA3. [ Resuh] To obtain a higher germination rate of seeds, the kiwifruit seeds were treated with GA3 at 2.5 g/L for 12 hours, and sterilized with 10% of sodium hypochlorite for 10 minutes, then the seeds were sowed in culture utensil with moistened filter paper and dark cultured. The germinated seeds were inoculated timely on the medium ( MS + 2 mg/L 6-BA + 0. 05 mg/L NAA + 30 g/L sucrose + 7 g/L agar power, pH 5.8) and cultured in tissue culture room, then we can get lots of ki- wifruit seedlings quickly in this way. Rapid propagation individually of kiwifruit seedling by using the bud-stem segments as explants can obtain lines with the same genotype. [ Conclusion] 'Xuxiang' and 'Bruno' seedlings can be used as kiwifruit rootstocks, thus rootstock' s factory production of kiwifruit can be realized.
